Slightly acidic electrolytic water has a good bactericidal effect on foodborne pathogenic bacteria,and was considered as a wide-spectrum and high-performance bactericide that is safety,high efficiency,and harmless to humans and the environment are increasingly applied in the food industry.However,there are few studies of the bactericidal effect of slightly acidic electrolytic water on Cronobacter sakazakii,and further research shoule be investigated.Therefore,this study was designed to evaluate the the bactericidal effect of slightly acidic electrolytic water on C.sakazakii and application in stainless steel and rubber surfaces.In this study,Cronobacter sakazakii ATCC 29544 was selected as research strains.The bactericide effect of the slightly acidic electrolytic water was evaluated by measuring the effect of different concentrations of the slightly acidic electrolytic water on C.sakazakii.The further bactericidal effect of the slightly acidic electrolytic water on C.sakazakii was investigated by measuring the intracellular ATP content,potassium ion leakage,protein leakage,intracellular p H,ROS content,DNA changes and cell morphology.In addition,C.sakazakii was inoculated on stainless steel and rubber surfaces to evaluate the bactericidal effect of slightly acidic electrolytic water on the surfaces.The results were as follows:The ability of bactericidal was increased with the concentration of the slightly acidic electrolytic water on C.sakazakii,and the number of colonies was decreased with the increase of the action time.C.sakazakii can be completely killed by 30 mg/L slightly acidic electrolytic water treated with 60S.The result found that the intracellular ATP content of the C.sakazakii cells was significantly decreased by measuring the ATP content of cells treated with slightly acidic electrolytic water,which indicated the C.sakazakii cells were damaged.By measuring the leakage of potassium ions and protein,it was found that the leakage of potassium ions and protein in C.sakazakii cells increased significantly after the treatment with slightly acidic electrolytic water?p<0.05?,indicating that the cells membrane might be damaged and the membrane permeability was changed.By measuring the change of intracellular p H(p Hin)of C.sakazakii cells after treated with slightly acidic electrolytic water,it was found that the p Hin was decreased?p<0.05?,indicating that the membrane of the cells was damaged and could not maintain the normal p Hin.By measuring the content changes of reactive oxygen species?ROS?in C.sakazakii cells after treated with slightly acidic electrolytic water,it was found that the ROS content in bacteria cells was gradually decreased and not accumulate,which indicated cell membrane was damaged,leading to intracellular ROS leakage.Agarose gel electrophoresis was used to determine the the DNA changes of C.sakazakii cells after treatment with slightly acidic electrolytic water,the results showed that DNA bands of cells gradually became shallow,or even disappeared,indicating that the cell membrane was damaged,leading to bacterial DNA degradation.Transmission electron microscopy was used to observe the the C.sakazakii cells cell morphology,the results showed that the structure and morphology of the bacterial cells treatedwith slightly acidic electrolytic water were changed,the cell wall and membrane were separated or damaged,the cell contents were leaked,and the bacterial cells were seriously damaged.In the application research,the different concentrations of slightly acidic electrolytic water was applied to the surface of stainless steel and rubber.The results showed that the bactericidal effect of C.sakazakii on the surface of stainless steel and rubber was increased with the concentration of the slightly acidic electrolytic water increasedly?p<0.05?,and the longer the action time,the better the bactericidal effect.In summary,the slightly acidic electrolytic water showed effectively bactericidal activity on C.sakazakii,and the bactericidal effect was increased with the increase of concentration and time.The slightly acidic electrolytic water can damage the cell membrane and lead to the change of ATP content,increased potassium ion leakage,increased protein leakage,decreased p Hin,decreased ROS content,DNA degradation and cell morphological changes,thus resulting in bacteria to be killed.In addition,C.sakazakii that was inoculated on stainless steel and rubber surfaces can be effectively decreased by slightly acidic electrolytic water.
