In the early stage of the project team,the in vitro antibacterial screening test showed that the Halostachys Caspica had obvious antibacterial effect against Staphylococcus aureus and Escherichia coli.Therefore,this experiment used Halostachys Caspica as the research material to evaluate its nutritional value,screen its antibacterial components,and determine its Antibacterial spectrum and antibacterial potency,separation of bacteriostatic active monomers,and bacteriostatic inhibition of rumen bacteria in vitro.The specific contents and conclusions are as follows:Test 1: Analysis of nutritional value of Halostachys Caspica in different periodsThe purpose of this experiment is to evaluate the nutritional value of the semi-desert land around the Aral City in Xinjiang and the Halostachys Caspica growing on the desert edge of the Taklimakan Desert.The crude nutrient analysis method is used to determine the salt panicles in the greening period,the vigorous growth period and the withering period.Halostachys Caspica samples were analyzed for nutrients.The results showed that the content of conventional nutrients in the three periods of Halostachys Caspica was different,the content of crude protein in semi-desert land was significantly higher than that in desert(P<0.05),and the content in re-greening period was significantly higher than other periods(P<0.05).During the period,the coarse ash content was significantly higher than that of semi-desert land(P<0.05);the ether extract content was significantly higher than other periods(P<0.05),the highest was 10.39%,and the content changed first.The trend of post-reduction;the difference of acid detergent fiber in three periods was not significant,but in the regreening period,the content of semi-desert was significantly higher than that of desert(P<0.05);the content of neutral detergent fiber was semi-desertly higher than that of desert.(P<0.05),the highest was 45.35%;the difference of calcium and phosphorus content was significant(P<0.05),but in the same period,there was no significant difference between desert and semi-desert land,calcium and phosphorus in The changes in the three periods showed a trend of increasing first and then decreasing.By contrast,in the semi-desert areas where the soil is more fertile,the nutritive value is higher during the period from green to vigorous growth,which can be used for the development of roughage.Test 2: Analysis of chemical constituents of Halostachys CaspicaThis experiment uses the system pre-test method to pre-test the chemical components contained in the Halostachys Caspica,and through the test tube reaction,the filter paper reaction,the color reaction of various indicators and color reagents,or the precipitation reaction.The liquid,95% ethanol extract andpetroleum ether extract were pre-tested for chemical composition,and the chemical species of Halostachys Caspica were initially explored.The results showed that the Halostachys Caspica contains amino acids,peptides,proteins,sugars,organic acids,saponins,alkaloids,volatile oils and oils,steroids or triterpenoids,flavonoids,phenols,tannins,and terpenoids.And steroidal lactone compounds and other components provide theoretical guidance for further study of the biological activity of Halostachys Caspica and its development and utilization.Test 3: Extraction,isolation and identification of antibacterial components from Halostachys CaspicaThe purpose of this test is to study the antibacterial monomers of Halostachys Caspica,crude extraction of Halostachys Caspica,separation and purification by column chromatography,activity determination of bacteriostasis test,monomer detection by thin layer chromatography,and GC / MS The monomers were identified and the bacteriostatic effects of alkaloid monomers were determined.The results showed that 26 components were separated by column chromatography.The n-butanol component had bacteriostatic activity.Among them,components 3-6 had the best antibacterial effect,followed by components 3-5.The structure of 3-5 is sophoroline,and the structure of monomer 3-6 is ephedrine.The minimum bacteriostatic concentration(MIC)of alkaloid monomer(ephedrine)of Halostachys Caspica is0.025 g / mL,and the minimum bactericidal concentration(MBC)is 0.05 g / mL,and the bacteriostatic effect is significant.Test 4: Analysis of inhibitory effect of bacteriostatic monomer from Halostachys Caspica on rumen microorganismsThe aim of the experiment was to study the inhibitory effect of alkaloids on the rumen bacteria,and the antibacterial activity was evaluated by an in vitro antibacterial test method.Six rumen bacteria were isolated from the rumen.By morphological observation,Gram stain and physiological and biochemical identification,bacteria A and B were identified as rumen genus,bacteria B and bacterium F were Lactobacillus,and bacteria D was short bacillus.Genus,bacteria E is the genus Bifidobacterium.The alkaloid monomer has the strongest inhibitory effect on D bacteria,the minimum inhibitory concentration is 0.025 g/mL,and the minimum inhibitory concentration against B bacteria,F bacteria and E bacteria is0.05 g/mL,and the inhibition effect on A and C bacteria is weak.The minimum inhibitory concentration is0.1 g/mL.It is indicated that the Halostachys Caspica alkaloid base has an inhibitory effect on rumen bacteria.
