Red yeast rice is a medicinal and food-grade rice koji.It is loved by people because of containing functional substances such as blood pressure lowering,blood fat lowering and blood sugar lowering.These active substances in red yeast rice are widely used.Although many metabolites have been isolated and identified,other natural products with antibacterial,anti-oxidant and anti-tumor activity still have some significance.In this paper,Fujian Gutian red yeast rice was taken as the object of study,and the similarities in the chemical composition of red yeast rice from Gutian and other regions,different organic solvent extracts from Gutian red yeast rice were also explored.At the same time,the antibacterial,antioxidation and cytotoxic activity in vitro of crude extracts of red yeast rice were studied.Based on this,the purification and structure identification of the active substances of the extracts of Gutian red yeast rice were carried out.Finally,the purified compounds were studied for antibacterial,antioxidative and antitumor activities.The main research results are as follows:?1?Liquid fingerprints of different regions red yeast rice and different organic solvent extracts from Gutian red yeast rice were established by optimizing HPLC chromatographic conditions.Similarity was calculated by the similarity evaluation system of fingerprint similarity,and it was found that the chemical compositions of Gutian red yeast rice differ greatly from the Zhejiang Lishui and Guangdong Meizhou at the wavelength of 302 nm.In addition,the chemical compositions of methanol extract differed significantly from ethyl acetate and petroleum ether extracts from Gutian red yeast rice.?2?Antibacterial experiment showed that the antibacterial effect of different organic solvent extracts of Fujian Gutian red yeast rice was significantly different.At the same concentration,the susceptibility of the various strains to the crude extracts of red yeast from strong to weak was bacillus subtilis>E.coli>aspergillus Niger>tower bin aspergillus>brewers yeast.Anti-oxidation experiments show that the Gutian red yeast rice methanol extract has stronger DPPH free radical scavenging activity than the other origins;Compared with other solvents,the ethyl acetate extract has the better antioxidant effect.Cell cytotoxicity assay showed that only acetone and ethanol extracts inhibited the proliferation of LO2 cells.The red yeast rice extracts can inhibit the proliferation of HepG2 cells to a certain extent and has a cytotoxic effect on HepG2cells.?3?A total of 12 compounds were isolated from Fujian Gutian red yeast rice methanol and petroleum ether extracts.They were determined by nuclear magnetic resonance,mass spectrometry and other spectroscopic methods,and the data were analyzed and compared with the database,compounds were identified respectively palmitic acid,linoleic acid,?-linolenic acid,methyl oleate,erucamide,triolein,oleic acid,diphenyl phthalate,erucic acid,methyl pentadecanoate,dibutyl phthalate and pregnenolone.?4?Antibacterial,antioxidant and cytotoxic activity in vitro experiments showed:at 40mg/m L,?-linolenic acid,dinonyl phthalate,dibutyl phthalate,linoleic acid,and triolein all have a certain antibacterial effect on Bacillus subtilis and E.coli.Alpha-linolenic acid,linoleic acid and oleic acid all have a certain ability to scavenge DPPH free radicals.In cytotoxic activity experiments,at the concentration range of 3.125-100.00?g/m L,palmitic acid,dibutyl phthalate,dinonyl phthalate,methyl pentadecanoate and alpha-linolenic acid all have a certain cytotoxic effect on proliferation of HepG2 cells.The results of this study can lay the foundation for the development of new drugs that have the effects of delaying aging and anti-tumor,and it provides a reference for the therapeutic and medicinal studies of natural products in red yeast rice.
