Sauerkraut is made from cabbage was taken as sample material,adding low concentration of salt water and using beneficial microorganisms on the surface of vegetables for fermentation.Sauerkraut is rich in nutrition and contains a variety of lactic acid bacteria.Northeast sauerkraut produces a variety of microorganisms in fermentation,and lactic acid bacteria is the main bacteria in the fermentation of sauerkraut.Strengthen the understanding of its biodiversity is important for the development of functional lactic acid bacteria resources in sauerkraut.This paper aims to explore the lactic acid bacteria diversity distribution in traditional northeast fermented foods,and the grid design idea was used to analyze and investigate the lactic acid bacteria resources in a single geographical source(Shuangyashan City,Heilongjiang Province).In this study,pheS(coding phenylalanine-t RNA synthase subunit)gene sequence analysis were used for identify species.The new strains of lactic acid bacteria were identified and their taxonomic status was determined.In this study,401strains of lactic acid bacteria were isolated from naturally fermented sauerkraut juice in Shuang ya shan,16S rRNA gene sequence analysis,pheS(coding phenylalanine-t RNA synthase subunit)gene sequence analysis and rpoA(coding RNA polymerase subunit)gene sequence analysis were used for identify unknown species.The results showed that 401 strains were distributed in 22 known species and 5 unknown species.It was distributed in 5 genera:Enterococcus,Lactobacillus,Pediococcus,Leuconostoc,Weissella.The 22 known species were:Enterococcus deviresei,Enterococcus xiangfangens,Lactobacillus brevis,Lactobacillus buchneri,Lactobacillus concavus,Lactobacillus coryniformis,Lactobacillus curvatus,Lactobacillus koreensis,Lactobacillus nenjiangensis,Lactobacillus nodensis,Lactobacillus parabuchneri,Lactobacillus paracasei,Lactobacillus paraplantarum,Lactobacillus plantarum,Lactobacillus sakei,Lactobacillus tucceti,Lactobacillus xiangfangensis,Lactobacillus daoliensis,Leuconostoc mesenteroides,Pediococcus inopinatus,Pediococcus parvulus,and Weissella paramesenteroides.Lactobacillus brevis and Lactobacillus plantarum were the dominant strains among the identified species,2 strains belong to Enterococcus,2 strains belong to Pediococcus,1 strain belong to Weissella,1 strain belong to Leuconostoc.In the pheS gene sequencing,unknown species 1represented by J23 P-F D08 in close phylogenetic branches to Lactobacillus nangangensis 381-7~T.In the pheS gene sequencing,unknown species 2 represented by J45 P-F F07 in close phylogenetic branches to Lactobacillus songbeiensis 398-2~T.In the pheS gene sequencing,unknown species 3 represented by B21 P-F B07 in close phylogenetic branches to Lactobacillus yonginensis THK-V8~T.In the pheS gene sequencing,unknown species 4 represented by A44 P-F A02 in close phylogenetic branches to Lactobacillus coryniformis subsp.coryniformis LMG 9196~T.The unknown specie 5 represented by 834-9 was related to the phylogeny of Lactobacillus jixianensis 1594~T and Lactobacillus camelliae LMG 24277~T in 16S rRNA sequence analysis.The similarity of 16S rRNA sequence are 96.7%and 98.3%respectively,and the similarity of pheS gene sequence are 82.3%and 81.0%,and the similarity of rpoA gene sequence are 85.8%and 87.3%,the main fatty acids C18:1?9c and C16:0.And multiple classification methods to determine the taxonomic status of 834-9.The type strains and corresponding names were as follows:Lactobacillus sp..In this study,a unknown specie of lactic acid bacteria was isolated from fermented sauerkraut in Shuang ya shan area,which expanded the existing classification system of lactic acid bacteria and enriched the classification system of lactic acid bacteria.The isolated and preserved lactic acid bacteria resources provide strain resources for the high quality industrial production of pickled cabbage.
