There are differences between individual cells.Even in the same growth environment,there are still some differences in cell morphology and gene expression.Studying the differences between individual cells has great significance for physiological pathology and early diagnosis of diseases.Traditional cell analysis methods can only obtain the average level of cell population information,while single cell analysis technology can obtain accurate information of cells at the individual level.Therefore,single cell element analysis plays an important role in revealing life activities.In this paper,inductively coupled plasma mass spectrometry(ICP-MS)was used as an analytical method to establish a single cell(SC)-ICP-MS elemental analysis method with high sensitivity and accuracy.The main research results are as follows:1.The effects of mass spectrometry parameters such as Dwell time(td)and settling time(ts)on the results of single particle analysis were compared.The gold nanoparticle reference materials(NIST 8012,NIST 8013,GBWE 120127)were analyzed..The results show that with a short dwell time(0.05 ms)and settling time(0 ms),a higher signal-to-noise ratio can be obtained and more nanoparticles are detected.The Au NP reference material was analyzed by this method,and the obtained particle size results were in agreement with the standard values.The detection limit of the number of gold nanoparticles in the method is 1.1×105L-1,and the detection limit of the particle size is 8nm.2.Establish a single cell analysis method,by analyzing the content of Yb in Raw264.7 mouse mononuclear macrophage leukemia cells,we compared Raw 264.7 mouse mononuclear macrophages at different exposure times.The phagocytosis of leukemia cells to the upturned material.3.Using a new type of dual-well silicon wafer that captures single cells while also adding space to the solution,we analyzed the silver in the single cell of Raw 264.7 mouse mononuclear macrophage leukemia cells by isotope dilution combined with LA-ICP-MS.The content of silver is obtained in a single cell.
