Objective: To analyze the relationship between long-chain non-coding RNA(lnc RNA)and diabetic nephropathy(DN),as the same time,to reveal the pathogenesis of diabetic nephropathy and provide clues for its treatment.Methods: 1.Animal experiments.A C57BL/6J mouse model of type 2 diabetic nephropathy was established.Carry out biochemical testing: the collected blood and urine samples are tested for blood sugar,proteinuria,urine creatinine and other biochemical indicators.Then proceed to pathological examination: use hematoxylin-eosin(H&E)and Masson to stain tissue sections to observe the changes in kidney tissue.Follow-up sequencing analysis is in progress.2.The GWAS data of lncrna related DN were analyzed by meta-analysis.Search NCBI and GWAS catalog database,and conduct meta analysis on GWAS data.3.Based on GEO database,mining lnc RNA data related to DN.Search the GEO database to obtain the raw data of lnc RNA high-throughput sequencing related to diabetic nephropathy.Data mining was conducted through chip-seq,RNA-seq and lnc RNA-analysis processes to find out the differentially expressed genes and their involved biological processes and regulatory pathways.4.Establish the DN—lnc RNA network and find the lnc RNAs that are strongly related to DN.Use the lnc RNA expression matrix obtained by data mining to construct the network,visualize it with Pajek,and mine the key lnc RNA that has a strong correlation with DN.Result: The biological mechanism and pathway of differentially expressed genes were obtained by data mining.The DN-lnc RNA network was constructed,and two lncrnas closely related to DN were identified: NONMMUG000513 and NONMMUG008364.Conclusion: Subsequent biological experiments and in-depth research on the above results,especially lnc RNA and its biological mechanisms and pathways,can be used to reveal the pathogenesis of diabetic nephropathy and provide new clues and ideas for its treatment.
