Compared with traditional flocculants,biological flocculants have the advantages of no secondary pollution,wide use,low cost,high efficiency and non-toxicity.Therefore,this experimental study uses the degradation products of straw cellulose as a bioflocculant medium to reduce the cost of bioflocculant preparation,and analyzes and optimizes the components of the bioflocculant medium.This has important theoretical and practical significance for the high value-added recycling of straw and the development and utilization of low-cost bioflocculants.This research is based on the resource utilization of cheap straw waste materials,and find suitable cheap media through experiments to replace traditional high-priced raw materials.Through multi-generation separation and transfer technology,CMC-Na hydrolysis circle measurement and filter paper degradation straw experiment were carried out to obtain 5 strains with obvious cellulose degradation ability,and a cellulose degradation composite bacterial agent was constructed Under 30? incubation conditions,after 10 days of reaction,the filter paper strips turned yellow and thin,almost degraded By identifying the composition and community properties of the straw-degrading microbial agent,the obtained microbial agent was constructed and named Bacillus sp JF1,and a 30-day straw degradation test was conducted.Through mixed bacteria fermentation,the culture test of flocculent bacteria was conducted Using cellulose-degrading bacteria to carry out corn straw degradation pretreatment experments,the degraded straw is used as the main carbon source in the fermentation substrate for the pretreatment of straw fermentation broth;the straw degradation product is used as a low-cost culture medium for flocculants,Cultivate flocculent bacteria,get flocculant culture solution(bacterial suspension,fermentation broth,supernatant),and determine the reducing sugar concentration of 13.73g/L.31.96g/L and 44.5g/L,respectively.The results showed that the content of reducing sugar in the supernatant of flocculent bacteria was higher than that of bacterial suspension and fermentation broth.The flocculation rates of the bacterial suspension,fermentation broth and gupenatant of flocculent bacteria were 42%,70%and 78%,respectively,indicating that the extracellular secreted substances of the strain produced flocculants.The pig farm sewage was used as the experimental sample,and the flocculent bacteria were obtained after separation and screening.Through the optimization of the reaction conditions,it is concluded that the optimal fermentation time for flocculent bacteria is 2d,the optimal temperature is 35?,and the inoculation amount of fermentation broth is 3%The flocculant reacts for 30 minutes at 80?.which has high heat resistance;it reacts at 90?,showing a downward trend.High-throughput analysis.The strain is dominated by Bacillus at the genus level,with an abundance of up to 95%.The microbial flocculant was scanned by ultraviolet spectroscopy,and the results showed that the flocculant contained no nucleic acid and protein.Infrared spectroscopy analysis found that the flocculant material is mainly acidic polysaccharides of-OH and-NH2COCH3,and the active groups mainly exist in the form of COO-.Through EDX element analysis,it was found that the atomic percentages of the five elements in C,N,O,K and Ca in flocculent A1 were 67.8%,6.95%,24.67%,0.06%and 0.52%,respectively.
