Onychodactylus zhaoermii belongs to Amphibia,Caudata,Hynobiidae,Onychodactylus.It is distributed in Xiuyan Anshan and Daheishan Liaoyang,Liaoning province,northeast China.This research adopts simplified genome sequencing technology of molecular markers SNPs of O.zhaoermii(16 samples,respectively from the four geographical units,respectively is the Xiuyan Zuogou,the Xiuyan Zhonggou,Daheishan Shuigou and Daheishan Changlingzi).In order to provide a theoretical basis for the protection of O.zhaoermii,16 species of O.zhaoermii were excavated for SNPs,and the genetic structure and genetic diversity of the population were comprehensively analyzed.Results a total of 22.3Gb of high-quality sequence data was obtained by sequencing,with an average of 1.3Gb for each sample.The total number of high-quality SNP sites obtained was 18338 after filtering with the conditions of depth2×,Miss0.5,and(MAF)> 0.05.Genetic diversity of high quality SNPs showed that the results of Pi analysis of heterozygosity and nucleotide diversity showed that the genetic diversity of Daheishan Liaoyang area was greater than that of Xiuyan Liaoning area.Moreover,through the calculation of molecular variance,we can obtain the population polymorphism uniformity of the four geographical units.There is no population difference,but only the variation or diversity among individuals.PCA results of population genetic structure showed that there was no obvious aggregation among the individuals of the four geographical populations and the distribution was scattered.Moreover,the analysis results of population STRUCTURE also showed that no obvious population STRUCTURE was formed among the four geographical units.The results obtained from phylogenetic tree are consistent with those obtained from PCA and population STRUCTURE analysis.Appear this kind of circumstance,may be associated with Onychodactylus living environment,Onychodactylus live at the source of mountain streams,these sources are from the underground river,so the underground river or the direction of the underground river is connected between two will determine whether there is a gene exchange between each other,but now has not been reported about this aspect,further study is needed.The following conclusions can be drawn from this experiment:(1)The success of thisexperiment provides a new method and means for the O.zhaoermii and even Onychodactylus to obtain high-throughput SNPs genetic markers,which has high application value and application prospect;(2)The genetic diversity analysis results of SNP marker analysis showed that the overall genetic diversity of O.zhaoermii in Liaoning Xiuyan was low,and the genetic diversity in Liaoyang Daheishan was higher than that in Liaoning Xiuyan;(3)The population genetic structure analysis showed that no obvious population structure was formed among the four geographical units of O.zhaoermii;(4)It is suggested to strengthen the protection of O.zhaoermii.Based on the study results of population genetics,it is suggested to build a O.zhaoermii protection community in xiuyan and daheishan of liaoning province.
