Background and Objective:Colorectal cancer is one of the common malignant tumors in China The morbidity and mortality have been increasing in recent years.Most patients are in the middle and late stages when they suffer the disease Metastasis and recurrence are the main causes of death.Currently,surgical treatment combined with radiotherapy and chemotherapy are the major traeatment.so early screening and diagnosis of colorectal cancer are particularly important.With the continuous development of molecular diagnostics,molecular markers are gradually used for early screening and diagnosis of malignant tumors,which have the characteristics of high sensitivity and specificity.A large number of studies have proved that Tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activating protein(YWHAE)participates in various physiological activities of cells and plays a certain role in tumorigenesis,development and metastasis.This article examines the exprssion and bioinformatics analysis of YWHAE in colorectal cancer.The role of YWHAE was explored in the tumorigenesis,development and metastasis of colorectal cancer by mining the relevant professional databases,aiming to provide new tumor markers and gene therapy targets for early clinical diagnosis of colorectal cancerMethod:This experiment involves the following three aspects:(1)Chromatography and mass spectrometry are used to screen differentially expressed proteins.(2)Consult a large number of related literatures and select YWHAE as the target protein for this experiment.Western Blot technology was used to prove the expression of YWHAE in cancer tissues and adjacent tissues of colorectal cancer patients.(3)Mining related professional databases,and using software to integrate the relevant information of YWHAE in the database to study the relationship between it and colorectal cancerResult:1.Mass spectrometry-chromatographic analysis showed that there were 68 differentially expressed proteins in colorectal cancer tissues and adjacent tissues,of which 42 proteins were up-regulated in tumor tissues and 26 proteins were down-regulated in tumor tissues.The protein YWHAE is one of the proteins up-regulated2.The results of Western Blot experiments proved that the expression of YWHAE in colorectal cancer tissues was higher than that in adjacent normal tissues,and it was an up-regulated protein.3.Analysis and integration of the Oncomine database revealed that the expression of YWHAE in colorectal cancer tissues was higher than that in adjacent tissues;The search of Cbioportal database shows that the mutation frequency of the YWHAE gene in colorectal cancer is only 2.7%.At the same time,the co-expressed genes of YWHAE were analyzed.According to the correlation,the top ten genes were as follows GLOD4,COPS3,CIQBP,EIF4A1,RPA1,PRPSAP2,MRM3,TSR1,NUP88,SCO1.At the same time,the analysis showed that the co-expressed gene TP53 was positively correlated with the level of YWHAE.By analyzing the COSMIC database,we can analyze YWHAE mutation types and their proportions.Among them,nonsense mutations account for the smallest proportion,accounting for 0.91%,and missense mutations account for the highest,reaching 17.52%.The proportion of other mutation types is smaller.Proportion range is 0%-6.04%.The analysis of ClueGO and GluePedia can obtain the interaction between co-expressed genes of YWHAE and their signaling pathways.Among them,the most important signal pathway is corpus callsum morphogenesis,accounting for 37.93%,followed by positive regulation of cholesterol efflux,accounting for 17.24%.By analyzing the STRING database,YWHAE and its subtypes(YWHAZ,YHWAH)in the 14-3-3 protein family,RAF1,and BRAF are closely relatedConclusion:1.YWHAE is up-regulated in colorectal cancer tissues,and its expression in cancer tissues is higher than that in adjacent tissues2.The gene encoding YWHAE has a low mutation rate in colorectal cancer,which is suitable for the research on molecular markers and gene expression.The gene is closely related to multiple transduction pathways of colorectal cancer and may be a new tumor marker and molecular target for treatment of colorectal cancer.And it plays a role in the occurrence,development and metastasis of colorectal cancer,which deserves further research and exploration.
