Background: Dendrite is very important for neuron signaling.Developmental defect of dendrite can lead to multiple nervous system diseases,such as autism.Celsr2 is an atypical cadherin in the cadherin family.Celsr2 may play a role in dendrite development besides its classical regulating functions.Explaining its regulation manner and mechanism is of great significance for exploring the pathogenesis of related diseases.Objective: To study the role of Celsr2 in dendrite development of hippocampal neurons in vitro.Methods: Celsr2 knock-out mice were used in the research.Neurons were prepared from embryo 17.5 fetus for culture.Total dendrite length and branch number were calculated for analysis.Live cell workstation were used for time-lapse experiment,which was aiming to explore the manners of Celsr2 regulating the dynamic process of dendrite development.In addition,we use Celsr2::HA mice to study Celsr2 subcellular location.Transfection and immunofluorecence techniques were used for spine and synapse analysis,respectively.Results: We found that Celsr2 mutation causes overextension of dendrites by the manner of regulating extension and retraction of dendrite dynamic progress.Celsr2 has no impact on the spine density and classification in vitro.Celsr2 expresses at preand post-synaptic portion and affects the location of synaptic proteins and the establishment of synapse.Conclusion: Celsr2 regulates dendrite development of hippocampal neurons and the formation of synapse in vitro.
