Wnt5a and Wnt5 b belong to Wnt family proteins which are very important for a variety of developmental processes.Wnt5 a is a key regulator of animal development.Mice embryos lacking Wnt5 a can't mature or die within a very short time after birth.Wnt5 a is also involved in cell function,homeostasis,etc.after birth.Wnt5 a has been widely studied in cancer,but there are only a few studies in nervous system.Recent studies have shown that Wnt5 a is involved in axon guidance,cerebellum development,midbrain dopaminergic neuron generation,maintenance of adult hippocampal dendritic stability.Wnt5 b plays a role in retinal development,but its function in the nervous system remains largely unknown.Defining the expression pattern of genes is the basis of functional study,so it is our priority to study the expression profiles of Wnt5 a and Wnt5 b and the phenotypes of Wnt5 a and Wnt5 b knockout mice.We used wild-type mice to examine the expression pattern of Wnt5 a and Wnt5 b in brain sections at different developmental time points using the high-sensitivity RNAscope kit.We performed immunohistochemistry staining and HE staining of brain sections or whole mount embryo from Wnt5 a and Wnt5 b knockout mice,to analyze general brain development and nerve fiber development.We also did behavioral analysis of Wnt5 b knockout mice.Our results showed the expression pattern of Wnt5 a and Wnt5 b in different developmental stages.The outgrowth of nerve fibers in early Wnt5 a deficient embryos is abnormal,and Wnt5 a and Wnt5 b knockouts also show defects in brain development.Wnt5 b knockout mice display higher activity and less anxiety in the elevated plus maze and open field test,and reduced motor coordinate and learning ability in the rotarod test.These results are crucial for future experimental design for conditional knockout mice and mechanism studies...
