Antibiotic residues from antibiotic zymophyte slags generated during production in pharmacy industry become an awkward solid waste and unreasonable processing method would result in environment pollution and ecology hazards as well as resource wastes.Among many processing methods of antibiotic zymophyte slags,microbial degradation has been the hot spot in related research area for its advantages of mild reaction conditions,low energy consumption and environmental protection.This thesis takes lincomycin zymophyte slags as research object and studies the harmless treatment process of slags by microbial degradation.This research will lay the research foundation for harmless treatment of antibiotic zymophyte slags in pharmacy industry.The paper complete the following aspects:Firstly,screening a high-effective degradation bacteria strain by directive breeding from sludge that long-term contaminated in lincomycin hydrochloride,doing sequence alignment through 16 S r DNA sequencing analysis and physiological-biochemical characteristic of this strain,it comes to a conclusion that this strain belongs to ochrobactrum tritici named KDSP-J0105.Secondly,the biodegradation mechanism of target strain had been clarified through mass spectra?HNMR and CNMR analysis,it draws the conclusion that the biodegradation product of KDSP-J0105 lincomycin hydrochloride is propyl pyroglutamic acid.Thirdly,the flask shaking fermentation techinques conditions of KDSP-J0105 were discussed.The results showed the carbon source was soluble starch,the nitrogen source was yeast extract,the initial p H was 7,the temperature was 30? and the rotating speed was 200r/min.Fourthly,then taking KDSP-J0105 as the target strain,the impacts of initial p H,temperature and shaker rotating speed on its degradation characteristic had been included through single factor experiment.The degradation rate at 8h of KDSP-J0105 strain could be reached 98% under the condition that the initial p H was 8,temperature was 40? and rotating speed of 150r/min.Fifthly,from the view of application,the conditions of cells immobilization of KDSP-J0105 had been studied.When the concentration of sodium alginate is 3% and calcium chloride is 2%,stirring rate is 300r/min,immobilization temperature is 35? andtime is 18 hours,the mechanical strength of immobilization particles reaches the highest and degradation effect of lincomycin hydrochloride.Applying the immobilization particles for lincomycin degradation-separation coupling process with characteristic of new expanding bed separation equipment,the degradation rate of lincomycin hydrochloride can reach 100% at 22 h and the degradation product is harmless.Therefore the harmless treatment for zymophyte slags in pharmacy industry has been initially achieved.Provide basis research for the formation of industrialization.
