Research And Development Of Sucrose Sulfate Metal Salts As Disinfectant,Antimicrobial Laundry Detergents And Antisepsis Ointments

With the improvement of human material life level,the demand of people to health and living environment is higher and higher,and antibacterial products gradually become daily necessities for people.In recent years,antibacterial materials industry in China has entered a faster phase of development and application.When silver ion and copper ion which have excellent antibacterial properties combine with organic and inorganic carriers,some composite antibacterial agents would been produced.These antibacterial agents are becoming the mainstream because of its high efficiency,low price and high stability.Firstly,the article introduces the development and research status of antibacterial agents at home and abroad,and expounds the classification,characteristics and application of antimicrobial agent.Meanwhile,the article summarizes current research development of sucrose and its products.In this project,taking a sucrose as a raw material,we designed and synthesied three sucrose sulfate metal salts with antimicrobial activity,including silver salt,copper salt and silver and copper salt respectively.Then their bioactivity to inhibite gram bacteria was determined.Furthermore,we prepred three medical ointment using these three sucrose sulfate metal salts and carried out some biological experiments,for example,promoting the union of trauma and burn skin for rat,test of acute toxicity for rabbit skin.In additon,we made some antibacterial chemical agents by adding the sucrose sulfate metal salts to the market abluent,then evaluated their antisepsis activity.First,the sucrose is abundant natural resources in Guangxi.sucrose sulfate sodium salt was gained by the sulfonation of sucrose with chlorosulfonic acid-pyridine and neutralization of sodium hydroxide solution.Then sucrose sulfate silver salt,sucrose sulfate copper salt and sucrose sulfate silver-copper salt were obtained by the ion exchange of ucrose sulfate sodium salt with silver nitrate or copper sulfate.The structures of compounds were evaluated by IR and UV,and the degree of esterification was determined with methode of barium sulfate weight,and content of silver ion or copper by using Volhard method or indirect iodometric titration.Afterwards,the experiment of inhibiting bacteria was carried out by using sucrose sulfate silver salt,sucrose sulfate copper salt and sucrose sulfate silver-copper as an inhibitors,and staphylococcus aureus,escherichia coli,Klebsiella pneumoniae,pseudomonas aeruginosa and Proteus vulgaris as bacteria of test.Compared with anti-microbial drug sulfadiazine silver,we discovered that the three sucrose sulfate metal salts were a better inhibitors against escherichia coli,Klebsiella pneumoniae and Proteus vulgaris than sulfadiazine silver.Moreover,the inhibiting effect of sucrose sulfate silver salt to pseudomonas aeruginosa and sucrose sulfate copper salt to staphylococcus aureus was also better than sulfadiazine silver.Then the MIC value of sucrose sulfate copper salt to staphylococcus aureus,escherichia coli,pseudomonas aeruginosa and Proteus vulgaris was deterined by double dilution method,its MIC value is 0.461 mg/mL.Further more,selecting pseudomonas aeruginosa as an object,we compared the antisepsis ability of sucrose sulfate metal salt with sucrose sulfate sodium salt,silver nitrate and copper sulfate.The results showed that the sucrose sulfate metal salts had an better antisepsis ability than their precursors.Whereafter,we prepared a series of antibacterial products,such as sucrose sulfate metal salts sterilization disinfectant,sucrose sulfate metal salts laundry detergents and sucrose sulfate metal salts ointments.Furthermore,different biological activity tests of these antibacterial products were carried out.The antisepsis ability of antibacterial laundry detergent diluent was compared with general laundry detergent diluent and the result displayed that it had a better antisepsis activity than general product.It means that the sucrose sulfate copper salt could contribute as an use of daily antisepsis chemicals.Meanwhile,the effects of three sucrose sulfate metal salts medical ointment to promote the union of trauma and burn skin for rat were evaluated by compared with silver sulfadiazine ointment,menstruum and blank group.The results showed that sucrose sulfate silver salt and sucrose sulfate silver-copper salt are better promotors to the union of trauma skin than silver sulfadiazine ointment,and sucrose sulfate silver salt and sucrose sulfate copper salt are better promotors to the union of burn skin,compared with positive control,menstruum and blank group,and the union time was shorted.Last,the acute toxicity of sucrose sulfate metal salts medical ointment to normal skin and damaged skin of rats,and the irritability testing to normal skin and damaged skin of rabbits were evaluated.The results showed that rats and rabbit were normal about spirit,appetite,urination or defecation,limbs activity,breathe and weight,also never appeared death and red swelling around unguent.The conclusion is that they are nontoxic.Then tissue section examination of the neogenesis skin from trauma and burn rats was performed.The texture pathology showed that the drug group possessed a normal skin construction and little lesion,but the menstruum group and blank group had more phenomenon of dermis congestion,inflammatory cells infiltration and individual abscess.In conclusion,we had synthesized some sucrose sulfate metal salts with antimicrobial activity of broad-spectrum,highly effective and safety by using sucrose as a starting material.We prepared some daily chemicals with higher antisepsis activity by combining the sucrose metal salts with laundry detergent,hand washing,cleanser essence.Furthermore,combining the astringency of sucrose sulfate with activity of antisepsis and anti-inflammation for silver and copper,a series of new antibacterial anti-inflammatory ointments possessing to promote the union of trauma and burn skin were prepared.
