Persimmons are rich in polyphenols which have been known to possess beneficial functions,such as antioxidation,preventing cardiovascular disease,improving immunity,antitumor,multidrug resistance reversal,antidiabetes and acceleration of alcohol decomposition.This study researches on creating the extraction,separation and purification,detection methods of phenolic compouds from the persimmon and investigating the antioxidant activity.The main research content and conclusions are as follows:(1)Qualitative analysis results by 2%ferric chloride solution colorimetry indicated that the extract of persimmon contains polyphenols.The extraction of total polyphenols from persimmon was determined by Folin-Ciocalteu Colorimetry.The optimum conditions of extraction were confirmed by twice orthogonal experimentsas follows:60%ethanol solution,ratio of liquid to solid 15:1 mL/g,55℃,30 min,the highest extraction rate of persimmon polyphenols was 3.0667 mg/g F.W.Through HPLC/HPLC-MS qualitative and quantitative analysis,it was concluded that the extract of persimmon contained GA、C、EC、CA,which accounted for 45.45%of total polyphenols.The content of the four were:GA>C>EC>CA.GA was the highest accounting for 68.34%of all the four and 31.06%of total polyphenols.There were no FA、CHA detected in the extract of persimmon.(2)Through static state experiments,HP2MGL with high absorption and desorption rate was chose to purify polyphenols by dynamic absorption and desorption experiment,the purity of polyphenols increased from 45.37%to 88.52%.Established the separation and purification method of phenolic acids by Sephadex LH-20、ODS column chromatography.Finally,four phenolic compouds were separated,GA,C,EC,CA,the purity were respectively 95.15%,95.04%,87.47%,85.0%(3)The extract of persimmon peel and pulp,GA and C which were obtained by the separation and purification,took on the strong radical scavenging activities against ABTS and DPPH radicals.
