The study found that microbial fermented L-tryptophan is a very complex metabolism process,involving the supply of precursor material,a downstream product feedback suppression etc.Therefore,Strict control of the process of fermentation has a great influence on the acid production of L-tryptophan and high-density.Regarding recombinant E.coli strain KC102 as a starting,there are some experimental studies as follows:1.Through single factor experiment to optimize recombinant E.coli KC102's culture conditions,screen several factors that can affect E.coli producing L-tryptophan,then do steepest ascent experiment and response surface software design,experimentally derived shake bottle optimum conditions.2.Doing filling material batch fermentation experiment on 5 L fully automatic fermentation tank,confirm the optimal cultivation mode of 5L tank filling material batch fermentation experiments.3.Downstream processing,and confirm the best way to extract L-tryptophan,than ultra high performance liquid chromatography(HPLC)method is used to calculate the product yield and purity.
