Lignocellulose is the world's largest reserves of renewable biomass resources,it has widely distributed,abundant source characteristics.Fossil fuel resources in facing non-renewable natural environment deteriorating situation and crisis,the use of inexpensive and convenient lignocellulose to produce ethanol and other liquid fuels and related chemical products,Can effectively alleviate the human face of the two major energy and environmental crisis,improve dependence on fossil fuels and reduce the damage to the natural environment and promote sustainable development of the global economy.We live in an environment with a lot of natural lignocellulosic carbon sources of microbes and use them secreted cellulases to degrade lignocellulose is a development and utilization of this renewable resource of reasonable and effective way.Filamentous fungi on the natural environment of lignocellulose degradation plays a very important role,plays an important role.Therefore,the composition of lignocellulosic enzymes and enzyme induction of expression of these two areas for future efficient use of lignocellulose resources has very important theoretical significance and application value.The first factor of lignocellulose cellulase gene expression is a carbon source,in nature,filamentous fungi lignocellulosic enzyme gene will be induced only in the presence of a substrate of cellulose conditions[1].However,cellulose is an insoluble substance itself,it can not directly induce the expression of the gene of lignocellulose,but to hydrolyze cellfilosic substrate by the fungus itself,the background level of expression of lignocellulose,so that the release of extracellular oligonucleotides sugars,soluble oligosaccharide entry into a cell will induce the expression of a large number of lignocellulose cellulase gene[2].In order to identify what kind of oligosaccharides play a role in inducing,so far a variety of oligosaccharides and their derivatives a lot of detail.The results showed that cellobiose,sophorose,lactose,these types of saccharides and their derivatives can lead to induce a variety of filamentous fungi secreting cellulase.But if the disaccharide substance as an inducer,the production cost is too high,is not conducive to induce cellulase synthesis of industrial production.So were looking for cheap inducer of cellulase production has become an important issue to be solved.The problem with high lignin content biomass materials,low lignin content biomass material without lignin content biomass material as Trichoderma reesei cellulase production induced by screening object,three biomass material composition analysis,and then use them as an inducer were induced by Trichoderma reesei cellulase production,inducing its enzyme components of different enzyme activity,extracellular protein content,compared saccharification ability.Explore reesei extracellular enzyme production of the best inducer,effectively improve theTrichoderma reesei extracellular cellulase enzyme activity.By fluorescence assisted carbohydrate electrophoresis(FACE)resolves without lignin content biomass material Trichoderma reesei enzyme production significantly increased reasons;by proteomics(ITRAQ)Characterization of lignin content-free raw materials from substance to adjust the protein level T.reesei extracellular enzyme protein content to achieve better enzyme composition.Lay the theoretical foundation for future depth of cellulase and industrial applications.This work includes the following sections:1.Different lignin content biomass material induced production of cellulase Trichoderma reesei.Were selected high lignin content biomass materials,low lignin content biomass material without lignin content biomass material as Trichoderma reesei cellulase production induced by screening object,three main types of biomass material composition analysis,and then use them as an inducer were induced by Trichoderma reesei cellulase production,their different enzyme components induced enzyme activity,extracellular protein content,compared saccharification ability.Explore reesei extracellular enzyme production was the best inducer inducer without lignin content,it can effectively improve the Trichoderma reesei extracellular cellulase enzyme activity.2.No lignin content biomass material Trichoderma reesei enzyme production significantly increased reason by fluorescence assisted carbohydrate electrophoresis(FACE)resolution.After the study found no lignin content has a better inducer induction induction Trichodenna reesei cellulase production process,we have no further studies confirm the lignin content of the biomass material to fungal enzyme production induced by universal applicability.To find no gentian disaccharides and trisaccharides wood can be used as cellulases and hemicellulases natural inducible factor,can induce Trichoderma reesei abundantly expressed at lower concentrations of cellulase and hemicellulase.This indicates that the formal induction of these natural factors induce yielding T.reesei cellulase.3.proteomics(ITRAQ)Characterization of novel protein levels induced from the compositions induced extracellular T.reesei cellulase production components more balanced.In the study without lignin content of more effective inducer improve reesei extracellular cellulase production reasons later.At the protein level,by proteomics(ITRAQ)also confirmed the induction of new compositions by inducing the production of extracellular T.reesei cellulase component proportions more balanced,cellulase enzymes exonuclease,hemicellulose enzyme amylase,a-glucanase,1,3-glucanase,pectinase extracellular ratio were raised,thereby achieving cellulase and hemicellulase ratio optimization,explains the novel inducer induced the enzymatic hydrolysis of biomass to improve the ability of the material reasons.
