Rhodamine is a fluorescent dye that is based on xanthene.Under non-alkaline conditions,rhodamine molecules are composed of a quinoid structure and have strong absorption and fluorescence in the visible light region.The maximum emission wavelength is generally between 500-700 nm,which can effectively avoid biological system background fluorescence.The interference occurs in the lactone structure in the alkaline environment.The spirolactone structure destroys the conjugated structure and the planar rigid structure of the molecule,so it has no fluorescence,and it is used in environmental chemistry,information science,fluorescent labeling,molecular biology,and pharmacology.Learning and so on.Rhodamine fluorescent dyes have advantages such as high fluorescence quantum yield,large molar extinction coefficient,longer excitation wavelength,and large usable range of pH.In this paper,two fluorescent probes were designed with Rhodamine as the parent:Hypochlorite fluorescent probe Rh-ClO and pH fluorescent probe.Hypochlorite anions are ubiquitous reactive molecules in the terminal disinfection system,inflammatory stress and immune system.Therefore,the rapid and visual monitoring of ClO~-in water and biological samples is very meaningful for the assessment of water quality safety and toxicity of pollutants.In the first part of this thesis,colorimetric and fluorescent probes(Rh-ClO)based on rhodamine B fluorophores and 2-thiophenecarbonyl hydrazide have been designed and successfully used for ClO~-detection in water samples and HeLa cells.After the addition of ClO~-,the color of the solution changed significantly,and we observed that it changed from colorless to pink.At the same time,bright red fluorescence could be observed under UV light(365 nm)excitation.Rh-ClO showed high selectivity and high sensitivity to ClO~-,and the detection limit calculated from fluorescence titration was 7?M.Its advantages include rapid response to ClO~-in 10seconds,and ClO~-in water can be successfully detected by Rh-ClO and its test paper.In addition,images of HeLa cells co-stained with Rh-ClO and Rh123 indicate that Rh-ClO has excellent and rapid cell membrane permeability and mitochondrial targeting.It has been confirmed that Rh-ClO will become a promising fluorescent probe that can rapidly track ClO~-in water samples and mitochondria of living cells.In the second part of the thesis,rhodamine was used as the parent and reacted with glycine methyl ester to produce intermediate products and reacted with four aldehydes to generate a series of target substances.Through pH titration test and reversibility test,it can be known that the four target compounds have a rapid response to pH and are not affected by other metal cations,that is,four kinds of pH fluorescent probes.
