Corydalis yanhusuo is an important traditional Chinese medicine.Its main efficacy is activating blood and analgesia.It is widely used in clinical application.With the increase of the demand of Corydalis yanhusuo in the market,there has been adulteration and counterfeiting of herbs.In order to ensure the safety and curative effect of Corydalis yanhusuo,the establishment of its quality control method is particularly important.Ambient ionization mass spectrometry is a high-throughput,fast and simple analytical method,which has been gradually applied to the quality control of traditional Chinese medicine.In this paper,the method of rapid identification and quality control of Corydalis yanhusuo was established by using ambient ionization mass spectrometry technology.The main contents of the thesis research are:The first chapter summarizes the main chemical constituents,pharmacological effects and quality control methods of Corydalis yanhusuo.The basic principle and application of ambient ionization mass spectrometry technology are also reviewed.At the same time,it briefly introduced the influence of Chinese medicine concoctions on the drug ingredients.In the second chapter,the extraction method and solvent of Corydalis yanhusuo were optimized.The HPLC fingerprint of Corydalis yanhusuo was established.The chemometrics software was used to statistically analyze the HPLC fingerprints of various batches of Corydalis yanhusuo.It was found that the chemical composition of genuine and fake products of Corydalis yanhusuo differs significantly.The similarity between genuine and common model is more than 0.8,and the similarity of counterfeit is lower than 0.2.In principal component analysis,the scores of PC1 and PC2 are 63.10%and 10.41%,respectively,which can be divided into two regions,which can achieve variety identification.Qualitative analysis of the chemical constituents in Corydalis yanhusuo by liquid chromatography mass spectrometry was carried out,confirming more than 10 chemical components.In the third chapter,the conditions of the paper spray mass spectrometry of Corydalis yanhusuo were optimized,and the AMS fingerprints of Corydalis yanhusuo were established.The AMS fingerprints of multiple batches of Corydalis yanhusuo were statistically analyzed with chemometrics software.The similarity between the original samples of Corydalis yanhusuo and the common pattern was greater than 0.75,and the similarity of counterfeit samples was lower than 0.38%PC1 and 9.92%of PC2,respectively,which was slightly higher than that of HPLC,and the rapid identification of Corydalis yanhusuo was realized.At the same time,the effect of metal modified paper base on AMS analysis of Corydalis yanhusuo was investigated.It was found that the response of alkaloids in Corydalis yanhusuo to metal paper was decreased when the metal modified paper was used for paper spray.The rapid identification of Corydalis yanhusuo can also be completed by different metal paper bases.In the fourth chapter,quantitative analysis of four alkaloids before and after processing of Corydalis yanhusuo was carried out by using HPLC.The linear correlation coefficients of protopine,tetrahydropalmatine,jatrorrhizine and palmatine were 0.99999,0.9998,0.99988 and 0.9997,respectively.The results showed that the content of tetrahydropalmatine,protopine,jatrorrhizine and palmatine in Corydalis yanhusuo increased.And the processing technology of Corydalis yanhusuo was optimized.At the same time,AMS fingerprints of unprocessed and processed Corydalis yanhusuo were established and combined with chemometrics software.The similarity of the common pattern between processed and unprocessed Corydalis yanhusuo was lower than 0.3.The scores of PC1 and PC2 were 77.73%and 9.95%,respectively.The rapid identification of processed products of unprocessed and processed Corydalis yanhusuo was completed.
