In recent years,fluorescent probes have been widely used in various fields such as biology and environmental science due to their advantages such as low cost,high selectivity,ability to visualize target regions,and good selectivity.The rhodamine lactam structure based on fluorescent probes has been widely reported.The pH-sensitive property of the fluorescent probe based on this structure also makes it often used in real-time monitoring of pH in biological cells,and has achieved good results.In this dissertation,rhodamine B was used as a fluorescent matrix.By modifying the structure of rhodamine B spirolactam,o-phenylenediamine series probes based on rhodamine B lactam structure and aminopyridine series probes were designed and synthesized.Among them,three probes with different recognition groups were tested for pH,metal ion interference,and reversibility in o-phenylenediamine series probes.After discussing and analyzing the properties of the three probes,it is believed that all three probes have the potential to become pH fluorescent probes.Aminopyridine series probes are divided into two groups: RAP series and RAPI series.In this paper,the two groups of probes were also compared,and a series of tests such as pH,metal ion interference,reversibility,solvent effect,and cell co-localization were performed.A mitochondrial pH fluorescent probe with good selectivity and anti-interference to hydrogen ions was obtained.Due to the design and adjustment of the pH value of the probe,we obtained two unreported pH insensitive probes,RAPI2 and RAPI3,which have certain research significance.
