Objective Through the construction of targeted nano metal polymer probe positioning, and adriamycin(DOX)and colchicine(COLC)effectively loaded in the material(FA-GCGLD-DOX/COLC),respectively in vitro and in vivo research on human hepatocellular carcinoma cell(HepG2)killing effect, and use a variety of methods of detection and analysis that provides data support for further research. Methods And the construction of drug delivery carrier FA-GCGLD-DOX/COLC successfully synthesized by the scanning electron microscope to observe the physical structure, the use of cell cultured human hepatoma cells(HepG2),the drug release carrier of FA-GCGLD-DOX/COLC,GCGLD-DOX/COLC and DOX/COLC and human hepatoma cells(HepG2)culture and to study its effect on killing people hepatocellular carcinoma cells in vitro, proliferation of HepG2 detection of drug release carrier MTT colorimetry after implantation were observed with confocal laser microscopy combined with cells,MRI was observed in FA-GCGLD-DOX/COLC,GCGLD-DOX/COLC positioning function and DOX/COLC in vitro and in vivo,by the end of the subcutaneous tumor formation in nude mice injected with FA-GCGLD-DOX/COLC,GCGLD-DOX DOX/COLC/COLC and observe the inhibitory effects on tumor. Results Cytological experiments showed that the inhibitory effect of FA-GCGLD-DOX/COLC,GCGLD-DOX/COLC and DOX/COLC on HepG2 cells was greater than GCGLD-DOX/COLC and DOX/COLC,the difference was statistically significant(P<0.05).MRI images showed that FA-GCGLD-DOX/COLC had a good location in tumor tissues. The tumor volume of FA-GCGLD-DOX/COLC group was smaller than that of GCGLD-DOX/COLC and DOX/COLC,and the difference was statistically significant(P<0.05).Conclusion FA-GCGLD-DOX/COLC has a good target to inhibit tumor and has little negative effect on organism.
