White Hypsizigus marmoreus(Peck)Bigelow belonged to Lyophyllaceae,Agaricales,Agaricomycetes,Basidiomycota and was an edible mushroom.Polysaccharide was an important active substance in white H.marmoreus which was related to the application of white H.marmoreus.Based on the pharmacological model,the immunological activities of polysaccharides from white H.marmoreus were measured.The structural characterization and bioactivity of homogeneous fractions were studied.The main results were as follows:(1)The crude polysaccharides WHN,WHB and WHP were extracted step by step from the fruiting bodies of white H.marmoreus.Six fragments named WHN1,WHN2,WHN3,WHB1,WHB2 and WHB3 were isolated from WHN and WHB by DEAE-Cellulose ion exchange chromatography.The monosaccharide composition analysis and immunomodulatory activity of the six components were studied.Results showed that WHN1 and WHB3 exhibited better immunomodulatory activity.(2)Sephacryl S-300 gel chromatography was used to separate WHN1-1 from WHN1.WHN1-1,WHB3 and WHP were identified as homogeneous components by HPLC.The average molecular weights of three fractions were 2.4 × 105 Da,2.3 × 106 Da and 2.1 × 106 Da.The monosaccharide composition analysis showed that WHN1-1 mainly composed of arabinose,mannose and galactose at the molar ratio of 1.1: 1.0: 3.1.WHB3 and WHP were glucan.(3)The backbone of WHN1-1consisted of(1?4)-?-D-Galp,(1?6)-?-D-Galp and(1?6)-?-D-Manp with the branched chains 1-?-L-Arap,(1?5)-?-L-Arap and(1?6)-?-D-Manp substituted at O-3 position of(1?6)-?-D-Manp.The backbone of WHB3 consisted of(1?6)-?-D-Glcp with the branched chains 1-?-D-Glcp substituted at O-3 position of(1?6)-?-D-Glcp.The backbone of WHP consists of(1?3)-?-D-Glcp with the branched chains 1-?-D-Glcp substituted at O-6 position of(1?3)-?-D-Glcp.(4)The immunomodulatory activity and anti-tumor activity of WHN1-1,WHB3 and WHP were detected.The maximum proliferation index of WHN1-1,WHB3 and WHP were 1.43,1.53 and 1.62 at the concentrations of 50 ?g/m L.The doses of polysaccharides were 200 ?g/m L,20 ?g/m L and 20 ?g/m L when the phagocytosis activity was optimum.The maximum releases of NO were 4.5 ?mol/m L,4.0 ?mol/m L and 3.0 ?mol/m L at the doses of 500 ?g/m L.In vitro antitumor analysis showed that three polysaccharides had good inhibition activity.The highest inhibition rates were 38.90%,38.18% and 25.99% at the concentrations of 500 ?g/m L,500 ?g/m L and 100 ?g/m L,respectively.(5)According to the results of UV-Vis spectrum,the condition for synthesis of Ag NPs was Ag+: WHP at the weight ratio of 1: 4.FT-IR and XRD analysis showed that the silver particles were interacted with the hydroxyl groups in polysaccharide successful.In vitro experiments,Ag NPs were stimulated macrophage RAW264.7 proliferation activity,increaseed the phagocytic activity and inhibited the proliferation of Hep G2 cell.
