Reaction process and the denitrification effect of Anaerobic Ammonium Oxidation?ANAMMOX?are extremely easily affected when organic matter added,because of the functional bacteria generation time is long,and has a harsh growing conditions.The effect of organics on ANAMMOX reaction was different in long and short term test.So how to reduce the rate of organic matter on the activity of anaerobic ammonia oxidation bacteria?AAOB?is the difficulties of the application of ANAMMOX.The Up-flow anaerobic sludge bed?UASB?reactor could intercept biomass efficiently and maintain the microbial diversity by using the three-phase separator.That has absolute advantages when starting the ANAMMOX reaction.This study analyzed the inoculation sludge and load increase period of mixed sludge in the reactor by using high throughput sequencing technology to improve the reactor of the load by improving the method to determine the concentration of substrate,and that were on the basis of the tests in the ANAMMOX reaction started successful y.Taking the sludge from training after the success of ANAMMOX sludge in UASB reactor in serum bottle for anaerobic batch test.By adding organic matter of different concentrations in the matrix to inspected the influence of the concentration of organic matter on ANAMMOX reaction.By analyzing the organic matter concentrations of ANAMMOX reaction denitrification efficiency,two kinds of organic matter concentration were found through its role in promoting and inhibitory effect is most obvious.And choosing the different time points within the bottle of sludge samples by using high throughput sequencing technology to explore the differences and similarities under different conditions in the microbial community structure.The long-term effects of organics on ANAMMOX reaction in UASB reactor were carried out,and the test sludge at different concentrations was tested for microbial flora structure.Here are the main conclusions according to short-term test results.?1?ANAMMOX reactor had been successful y started up within 17 days,the up-flow anaerobic sludge blanket?UASB?reactor seeding with mixed anaerobic sludge of laboratory cultures of ANAMMOX function and aerobic activated sludge from a municipal sewage treatment plant in volume ratio 1:2.This processes could be divided into phases of hydrolysis,enhanced and steady.Anaerobic ammonium oxidation bacteria?AAOB?was enriched by improving the reactor volume load gradually after the steady.When the volume load increased from 0.10 kg·?m3·d?-11 to 0.44 kg·?m3·d?-1,the removing load of total nitrogen?TN?also increased from 0.09 kg·?m3·d?-11 to 0.42 kg·?m3·d?-1.The color of sludge changed by shal ow red and deepened gradually in the UASB reactor,and proportion of the sludge particle size is greater than 0.2 mm from 10.90%to 38.37%at this time.The sludge of inoculation and volume load increased phase was detected by high-throughput sequencing.It resulted that the Chloroflexi,Proteobacteria,WWE3,Actinobacteria,Planctomycetes and so on were the dominant species.The proportion of the Proteobacteria of denitrification bacteria was gradually reduced from 21.60%to 14.20%with the increase of AAOB enrichment degree,while the Planctomycetes was increased from 0.73%to 15.50%.Candidatus Brocadia,Candidatus Jettenia,Candidatus Kuenenia is the main species of Planctomycetes when volume load increased to 0.44 kg·?m3·d?-1 of reactor,and the Candidatus Brocadia is the main species of AAOB,which accounted for 13.40%.?2?When the organic matter of the ANAMMOX reaction were glucose,acetic acid and sodium acetate,concentration of COD were 20,80,150,250 mg?L-1 were have a certain role in promoting the ANAMMOX reaction.While phenol and methanol were have certain stimulation or inhibition on each test concentrations through the anaerobic serum bottle test.The concentration of COD was 150 mg?L-1 of sodium acetate promoted the ANAMMOX reaction most among these cases,the removal rate of ammonia and TN were 93.45%and86.35%after the test start of 4 h.The concentration of COD was 250mg?L-1 of methanol had the strongest inhibitory effect,and effection experienced activity to stimulate and restrain phase.Through analyzing the sludge samples when time is 0 h,4 h,14 h and 46 h in these cases by high-throughput sequencing,it was found that the variation of ANAMMOX bacteria in this system structure is smal er under the action of mmonium acetate than the methanol.The main microbes in the systems were Planctomycetes,Proteobacteria,Chloroflexi and Firmicutes.Candidatus Brocadia and Candidatus Kuenenia accounted for the largest ratio of the Planctomycetes,Denitratisoma and Limnobacter accounted for the largest ratio of the Proteobacteria.?3?By adding concentration 0,40,60,80 mg?L-11 sodium acetate in turn to UASB continuous flow reactor,the results show that ANAMMOX was not significantly affected under the concentration of COD was 20 mg?L-1,and the removal rate of TN and COD were higher than without adding organic matterr,respectively were 88.52%and 75.25%.When concentration of COD was 60 or 80mg?L-1,the different degree of inhibition affected ANAMMOX.In particular,removal rate of TN fell to 50.02%when concentration of COD was 80mg?L-1.By testing different concentration of organic matter in sludge samples with high-throughput sequencing,the biological communities had the minimum complexity under the condition of the concentration of COD was 40mg?L-1.In all the samples,Chloroflexi,Planctomycetes,Proteobacteria and Actinobacteria take more proportion.The Planctomycetes gradually decrease from 24.60%to 7.70%with the concentration increased of organic matter,and the Candidatus Brocadia of which decrease from 12.14%to 3.63%.The proportion of Proteobacteria increased from15.40%to36.30%when the concentration of COD changed from 0 to 40mg?L-1,and the Bdelloribrio of which had the biggest increase,its proportion increased from 0.01%to 8.39%.
