Micro-nano fiber is a kind of optical waveguide.Because of the low loss in optical transmission, strong evanescent field and the simple making process,micro-nano fiber has been widely used in optical fiber sensing. As an important means for protein separation and detection, polyacrylamide electrophoresis has been commonly used in trace mixture separating.Integrating the polyacrylamide gel electrophoresis and tapered micro-nano fiber sensor is helpful for the rapid separation and detection of protein, and it is also one of the important application of biomedicine detection. At present, there are many methods for detecting proteins, however, the process of detecting protein separation by using micro-nano fiber sensor has not been attempted. In this essay,a series of experiments have been designed to verify the possibility of detecting protein by electrophoresis of micro-nano fiber sensor. It is a good starting point for the combination of micro-nano fiber sensor and electrophoresis separation, and has a far-reaching application prospect in the future trace material detection. This essay combined the sensing characteristics of tapered micro-nano fiber sensor and the advantages of electrophoresis in protein separation and detection, and the creative realization of the on-line dynamic detection method of protein. The thesis mainly includes three aspects: the preparation of tapered micro-nano fiber sensor and research on its properties, the preparation of polyacrylamide gel, curing test, and on-line dynamic detection in the electrophoretic separation of proteins by tapered micro-nano optical fiber sensor, discussed and analyzed the experimental process and experimental results in the last. This work will have a positive impact and significant significance in the field of biomedicine and other detection areas.
