In the current market,the demand for chiral alcohol is relatively large,and the toxicity to aldehydes and ketones inside the body is relatively large.Therefore,aldosterone reductase has great research value.Aldecanone reductase is an enzyme that catalyzes the asymmetric reduction of the corresponding chiral alcohol with a prochiral carbonyl compound in the presence of NADH.These enzymes generally exhibit high stereoselectivity for a wide range of substrates under mild reaction conditions.Due to its excellent selectivity and environmental compatibility,aldosterone reductase has been widely used in the synthesis of industrial drugs in recent years.Saccharomyces cerevisiae aldehyde reductase GRE2 protein is a stress protein.GRE2 is an oxidoreductase that can reduce aldehydes and ketones,and can reduce the accumulation of aldehydes and ketones.Glucose dehydrogenase(GDH)is a member of the short-chain alcohol dehydrogenase family and consists of four identical subunits.It catalyzes the oxidation of ?-D-glucose to oxidize its carbonyl group to alcohol.It reduces the coenzyme I(The protons on NADH)or reduced nicotinamide adenine dinucleotide phosphate(NADPH)are hydrogen-transferred to the carbonyl group,leaving nicotinamide adenine dinucleotide(NAD +)or oxidized nicotinamide adenine dinucleotide phosphate.(NADP+),which has dual cofactor specificity.Using this feature,it is combined with GRE2 to form a cyclic reaction.GRE2 uses the coenzyme NADPH to reduce aldehydes and ketones to alcohol,which in turn reduces NADP+ to NADPH,which can be recycled in the reaction.In this article I have constructed three plasmids for the GRE2 and GDH systems,GRE2 and GDH,respectively,on an intein-containing vector,loaded on a single-enzyme vector,with GRE2 and GDH,respectively.They were separately mounted on a single-enzyme vector containing a polypeptide,and the enzyme activity,enzyme stability,and secondary structure of the three systems were explored.Double-enzyme systems with inteins and peptides have been found to have better enzyme activity and stability than mixed single enzymes.It can be used in industrial production in the later period.
