Chinese giant salamander?Andris davidianus?is a kind of traditional rare medicinal animal in China,which has high nutritional value and has the effects of nourishing yin,tonifying kidney,enriching blood and promoting the circulation for vital energy.With the maturity and popularization of large-scale domestication technology for Chinese giant salamander,Chinese giant salamander industry has gradually become the most dynamic emerging industry to adjust the agricultural industrial structure and promote farmers income.The content of unsaturate fatty acid?UFA?in that Chinese giant salamander oil is high,?66.05-69.81%?,which has various pharmacological effects of preventing myocardial infarction,reducing blood fat,resisting oxidation and anti-aging.Thus,it has great development and utilization value.The antioxidant activity of Chinese giant salamander oil in vitro,the material and proportion of Chinese giant salamander oil microcapsules,the optimization of preparation process,physicochemical properties and storage stability of Chinese giant salamander oil microcapsules were studied in order to provide the basis for the development of Chinese giant salamander oil products,which can actively promote the deep processing of Chinese giant salamander.Antioxidant activity of Chinese giant salamander oil was tested by antioxidant experiment in vitro.DPPH radical,hydroxyl radical,superoxide radical scavenging ability,reducing power and Fe2+chelating ability of Chinese giant salamander oil were determined,to evaluate the antioxidant activity of Chinese giant salamander oil in vitro.The results showed that the IC500 of DPPH radical,hydroxyl radical and Fe2+chelating ability of Chinese giant salamander oil was 3.523 mg/mL,0.493 mg/mL and 0.559mg/mL,respectively.the IC500 of Chinese giant salamander oil added with antioxidant was 0.572 mg/mL,0.099 mg/mL and 0.062 mg/mL,respectively.The antioxidant capacity of Chinese giant salamander oil was stronger than that of mackerel oil.The antioxidant capacity of Chinese giant salamander oil was stronger than that of mackerel oil.The chelating ability of Chinese giant salamander oil to Fe2+was the best.Chinese giant salamander oil was stronger than Vc at the same concentration.The scavenging and reducing ability of giant salamander oil to DPPH radical and hydroxyl radical were equal to Vc.In order to ensure the stability of Chinese giant salamander oil,its microcapsules were prepared by spray drying method.Firstly,the preparation material and the proportion was screened.After selecting wall materials,the effects of wall material ratio,wall core material ratio,wall material content and emulsifier content on microencapsulation of Chinese giant salamander oil were studied with embedding rate as index.On the basis of 2%emulsifier addition,the response surface software box-behnken design model was used to determine the optimal proportion of Chinese giant salamander oil microencapsulation materials.The results showed that the ratio of wall material to core material was 3.44:1,the ratio of wall material to core material was5.37:1,and the amount of wall material was 6.46%.under the optimal conditions,the microencapsulation rate was 88.06%.In order to improve the encapsulation efficiency of Chinese giant salamander oil microcapsules,spray drying process parameters were optimized,the effects of inlet temperature,feed rate,homogenization time and homogenization speed on the encapsulation efficiency of Chinese giant salamander oil microcapsules were studied.The optimum preparation technology of Chinese giant salamander oil microcapsules was determined by orthogonal optimization design experiment.The results showed that the embedding rate of Chinese giant salamander oil microcapsules increased to 94.73%at the inlet temperature of 190?,the feed rate of 3.5 mL/min,the homogenization time of 9 min and the homogenization speed of 10000 r/min.The physical and chemical properties of Chinese giant salamander oil microcapsules were determined,and its stability was also studied.The water content of the microcapsule obtained by spray drying is 3.05±0.04%,the angle of repose of the microcapsule is 35.41±0.37°?30°<angle of repose<45°?,the density is 0.28 g/cm3,the particle size is 5-10?m,it has small viscosity regular shape,smooth surface,good fluidity and it quality is good.In addition,the storage stability of Chinese giant salamander oil microcapsules and the storage conditions of microcapsules in different storage environments?temperature,light,oxygen?were investigated with peroxide value?POV?and storage rate as indicators.The results showed that microencapsulation of Chinese giant salamander oil could prolong the shelf life of Chinese giant salamander oil.The microcapsule POV is higher at 25?than 4?,aerobic condition is higher than anaerobic condition,illuminated condition is higher than lightless condition.In addition,the storage rates of oil,DHA and EPA in Chinese giant salamander oil microcapsules were higher at 4?than at 25?,it was higher in anaerobic conditions than in arerobic conditions,and it was higher at lightless conditiot than at illuminated condition.Therefore,microcapsules should be stored in low-temperature vacuum light-proof conditions.
