Gastric cancer is one of cancer with high rates of morbidity and mortality.In China,gastric cancer is the most common cancer except for lung cancer and breast cancer.And so far,there are no other effective treatments other than surgery clinically.Therefore,there is still much potentiality for drug research on gastric cancer.A large number of studies have shown that isothiocyanate(ITCs)compounds have good anti-cancer effects,sulforaphane(SFA)and sulforaphene(SFE)are two structurally similar natural extracts of isothiocyanates which were found could inhibit proliferation in breast cancer,esophageal cancer,and non-small cell lung cancer in previous studies.Our research concentrated on gastric cancer and evaluated anti-gastric activity of sulforaphene and sulforaphane and molecular mechanism.The results are as follows:1.The inhibition effect of sulforaphene and sulforaphane on proliferation of gastric cancer cells has a concentration-dependent and time-dependent manner.In addition,sulforaphene and sulforaphane have the effects of inhibiting the cell cycle of gastric cancer cells and promoting apoptosis.And in animal experiments,sulforaphene and sulforaphane also have good anti-cancer effect.2.Sulforaphane has a good inhibition effect on the invasion and metastasis of gastric cancer cells.And through RNA-seq,we found sulforaphane's anti-gastric cancer target gene ESRP2.Moreover,the relevance of sulforaphane and its target gene ESRP2 with gastric cancer cell Epithelial-Mesenchymal Transition(EMT)pathway had been found.3.Through RNA-seq,we discovered ADGRA2,a potential target gene of sulforaphene,and initially explored the relevance between the target gene ADGRA2 and the Wnt/B-catenin pathway.
