This article is based on the crown pear cultivated in wuwei city gansu provinve.Respectively using different extraction parameters on the crown of chlorogenic acid in the pear skin slag optimized extraction,and compare the three different kinds of extraction process of chlorogenic acid in pear residue extraction effect,effect evaluation is extracted by extraction rate as index,find out the optimum parameters of the extraction process of chlorogenic acid in skin slag,finally in the research of crown pear skin slag the best extraction process of chlorogenic acid,using macroporous resin method for the extraction,isolation and purification of chlorogenic acid in the use of purified chlorogenic acid to remove DPPH · and · OH free radical,and by using spectrophotometry to determine the effect of free radical clearance,to judge from the crown pear skin slag to extract the antioxidant activity of chlorogenic acid.The results are as follows:1.Refer to the material,In order to first determine the content of chlorogenic acid by ultraviolet spectrophotometry,and the maximum absorption wavelength of chlorogenic acid was selected to be 330 nm.2.Application of single-factor test and orthogonal test design research using ultrasonic method extracted the optimal extraction process parameters of chlorogenic acid from crown pear slag.Influencing the ultrasonic extraction of chlorogenic acid in the crown pear skin slag for primary and secondary factors is that solid-liquid ratio > extraction time > extraction temperature > extracting agent concentration,the best extraction technology conditions for chlorogenic acid: 50%as extraction solvent,material liquid proportion is 1:25,the extracting time is 30 minutes,extraction temperature 40?,in the process conditions,the extraction rate of chlorogenic acid was 2.121%.3.The optimum extraction process parameters of chlorogenic acid in pear slag were studied by single factor test and orthogonal experiment design.The experiment results showed that the main factor of the four factors influencing the microwave leaching of the crown pear skin residue was:The concentration of the extract > microwave power > extraction time >material-liquid ratio.Crown pear skin slag by microwave assisted extraction of chlorogenic acid in the best process conditions for: material liquid ratio: 1,under the condition of microwave power 200 W,extracting time: 15 min,the extraction temperature 40?,extracting 3 times,under the condition of the extraction rate of chlorogenic acid was 1.982%.4.The optimum technological parameters of chlorogenic acid in crown pear residue were extracted by using single factor orthogonal experiment.The experiment results showed that the main factor of the four factors influencing the extraction of chlorogenic acid from pear slag was:The concentration of the extract>Water bath time>the extraction time>The solid-liquid ratio.Using solvent extraction to extract the crown pear skin residue of chlorogenic acid in the optimal process parameters for: material liquid ratio: 1:10,extracting agent: 70% methanol,water bath temperature: 50?,the extraction time is 3 h,extracting 3 times,under this condition,the extraction yield of chlorogenic acid was 1.363%.5.use NKA ? type of macroporous adsorption resin crown pear skin residue of chlorogenic acid in the static adsorption and desorption performance,speed and elution on the samples and process parameters are studied,identified by NKA ? type macroporous adsorption resin purification crown pear skin residue of chlorogenic acid in methods and conditions,and the high purity of chlorogenic acid.6.Using the Vc standard as the control,the removal ability of the purified chlorogenic acid to DPPH free radical and the free radical was determined by using the visible spectrophotometry method.The results showed that the removal ability of chlorogenic acid to DPPH and OH free radical was higher than the Vc clearance rate of 90% and over 20% respectively.And the clearance ability and content are in a certain amount of relationship,the higher the concentration,the stronger the clearance ability.
