Polymer microspheres refer to the spherical nano-structure with diameters ranging from nanometer to micrometer.Polymer microspheres have many outstanding merits,including large surface area,volume effect and good biocompatibility et al.These properties lay the foundation for the application of polymer microspheres in the fields of drug delivery,biosensing and cosmetics.However,due to their cumbersome preparation processes.The applications for drug carriers have't been used broadly.In this work,a new method for preparing polymer microspheres with simple operation and mild conditions has been developed.The prepared polymer microspheres are suitable to be used as drug carrier for cancer therapy.Nine kinds of poly?2-oxazoline??POx?with different side chains and different molecular weights were synthesized by cationic ring-opening polymerization.The polymer microspheres were prepared by hydrogen bonding between tannic acid?TA?and water or ethanol.The diameter of polymer microspheres can be adjusted by changing the molecular weight,concentration of POx and the ratio of POx and TA.The PMeOx2-TA/H2O microspheres with 90 nm diameter can be obtained,which is suitable to be used as a drug carrier.With the polymeric PMeOx2-TA/H2O microspheres in hand,doxorubicin PMeOx2-TA/H2O microspheres were prepared and a series of characterizations were performed.The UV-vis spectrometer?UV-vis?showed that the maximum absorbance of the doxorubicin-loaded microsphere located at 490 nm;The hydration particle size of the drug-loaded microsphereis about 100 nm from the dynamic light scattering?DLS?;The cytotoxicity experiment showed PMeOx2/TA has low biological toxicity;Confocal laser scanning electron microscopy?CLSM?was then used to verify that drug-loaded nanocomplexes can enter into the cell by the way of cell encapsulation.
