Mass spectrometer is not only an important analytical instrument,but also an important tool for studying chemical reaction.In mass spectrometry,gas-phase ion is mostly existed as a single molecule state,and ion/ion reaction can be highly effective.In addition,mass spectrometer can select specific ions to take reaction,and provide mass charge ratio information of the ions to distinguish products and reactant.Furthermore,through a variety of conversion methods such as tandem mass spectrometry,mass spectrometer can analyze the structure of the compound.In a word,mass spectrometer is an ideal choice for the study of gas phase ion / ion reactions.In order to determine the primary structure of a protein,it is primary important to reduce the disulfide bond in protein,and make it become a linear polypeptide chain.In this work,gas-phase ion / ion reaction was used for reducing the disulfide bonds in peptides and proteins with mass spectrometry.By this method,peptide with intermolecular and intramolecular disulfide bonds have been reduced.In addition,the ultraviolet photodissociation has also been proved to reduce the disulfide bond.
