High performance liquid chromatography?HPLC?is an extensively used separation and analysis method based on classic column chromatography.HPLC has been widely used in many fields such as medical health inspection,environmental analysis,agricultural drug analysis and food safety because of its advantages,including fast analysis speed,easy operation,high sensitivity and good reproducibility.Since the 1970s,separation of chiral compounds by HPLC has made remarkable progress in the field of chiral drugs.In this paper,chiral porous materials were used as chiral stationary phases?CSPs?in HPLC for separation of chiral compounds.The specific research contents are as follows:The first chapter mainly introduces chiral compounds,chiral separation,HPLC method,the resolution mechanism and types of HPLC chiral stationary phase,and the application of chiral porous materials in chiral recognition.A homochiral MOF[Cu3?HL?2?L?2?bpy?3]·4H2O was synthesized and used as chiral stationary phase for resolution of chiral compounds in the second part.The results showed that the CSP has a good separation ability toward 10 racemates.ThemonodispersechiralMOF@SiO2core-shellmicrosphereswas synthesizedused and used as CSPs for HPLC.The resolution ability of chiral MOF[Co2?D-cam?2?TMDPy?]@SiO2 column was investigated.There are 20 racemic compounds and 5 positional isomers were well separated on the column,and 12racemic compounds of them achieved baseline separation.The separation performance of the chiral column for racemic compounds under various factors including injection volume,mobile phase composition,the flow rate of mobile phase and column temperature was investigated.The reproducibility and stability of the chiral column were evaluated.The results indicated that the monodisperse chiral MOF@SiO2 core-shell composites as HPLC chiral stationary phase greatly improved the column efficiency and chiral resolution ability of the chiral MOF column.In the fourth part,a chiral metal organic cage[Fe4L6]?ClO4?8·solvent was synthesized,which was used as a novel CSP to prepare HPLC chiral column for separation of chiral compounds.The experimental results showed that chiral metal organic cage[Fe4L6]?ClO4?8·Solvent exhibited good resolution ability for 5 chiral compounds.In the fifth part,a chiral pentyl cage was synthesized and coated on the aminated silica gel.The separation performance of the pentyl cage CSP was investigated.The results showed that the chiral pentyl cage chiral column exhibited a certain ability for separation of five chiral compounds.
