Squid liver is rich in polyunsaturated fatty acids,such as EPA and DHA,and has many physiological functions,such as anti-inflammatory,anti-tumor,anti-thrombosis and so on.In this paper,the basic physical and chemical indexes of squid liver fish oil were determined to evaluate the quality of fish oil.The scavenging ability of free radicals was determined in vitro chemical system.The models of aging mice induced by D-galactose and acute liver injury induced by CCl4were constructed in vivo.The activity of related enzymes in serum of model mice was measured and the changes of histopathological sections were observed to evaluate the effect of squid liver fish oil on anti-aging and preventing liver injury.The contents of serum cytokines and their gene expression at the level of mRNA and protein were determined to reveal the functional mechanism of fish oil in the liver of squid from the molecular biological level.The research results as follows:?1?Measured the basic physical and chemical indicators of squid liver oil,with GC-MS method was developed for the determination of free fatty acids in fish oil composition,determined the squid liver oil in vitro antioxidant capacity in the pure chemical system.Experimental results show that the moisture and volatile matter,peroxide value,iodine value,saponification value indexes reach the standard fish oil secondary,the quality is good.There were many kinds of free fatty acids in squid liver fish oil,the content of polysaturated fatty acids reached 42.16%,among which EPA and DHA were 23.53%;Squid liver fish oil is rich in polyunsaturated fatty acids and has strong antioxidant ability in vitro.?2?By building D-galactose induced aging model mice,served the viscera index,the liver and skin pathological,the content of SOD,CAT and MDA in serum and the activity of GSH-PX and T-AOC,the content of serum cytokine IL-1?,IL-6,IL-12,TNF-?and the SOD1,SOD2,CAT,GSH in the expression of mRNA and protein level as an index in mice to evaluated the comprehensive of the squid liver oil of D-galactose induced aging mice model of antioxidant effect.The results showed that squid liver fish oil could alleviate the decrease of heart,liver,spleen and kidney index and delay the degeneration of viscera and organs in mice to some extent,increase the activity of GSH-PX and T-AOC and decrease the production of MDA in mice compared with the model group.In addition,fish oil in squid liver is rich in polyunsaturated fatty acids,which can reduce the release of inflammatory cytokines by reducing oxidative stress.Compared with the model group,squid liver fish oil could significantly reduce the content of inflammatory cytokine IL-1?,IL-6,IL-12,TNF-?,and increase the expression of SOD1,SOD2 CAT and GSH at the level of mRNA and protein,and reveal the mechanism of anti-senescence of squid liver fish oil from molecular biological level.?3?By constructed model with CCl4 induced acute liver injury in mice,served the mice viscera index,hepatic pathological,the content of ALT,AST,TG,GSH-PX and MDA in serum and the cell inflammatory factor IFN-?,IL-6,IL-12,TNF-?,and the expression level of the mRNA and protein of COX-2?iNOS?NF-?B and I?Ba in liver tissue as indexes to evaluated the effect of the squid liver oil to prevent CCl4 induced acute liver injury in mice.The results showed that squid liver fish oil could alleviate the increase of liver,spleen and kidney index and improve the pathological symptoms of liver in mice.Compared with the model group,the serum levels of IL-6IL-12 and TNF-?in serum of AST,ALT,MDA,TG and TNF-?were significantly decreased,and the content of TNF-?was increased.The polyunsaturated fatty acids in squid liver fish oil can reduce the chemotaxis of polynuclear cells and neutrophils to chemoattractant?such as leukotriene B4?,reduce the number of cell migration and inhibit inflammation.EPA and DHA could effectively inhibit the phosphorylation of NF-?B p65 pathway and decrease the expression of inflammatory factors.In addition,EPA and DHA could effectively prevent the degradation of I?Ba and inhibit the phosphorylation of NF-?B p65 pathway at the level of mRNA and protein.It can significantly reduce the expression of COX-2 iNOS in liver tissue,has a good anti-inflammatory effect,and has a certain preventive effect on liver injury.
