Seaweeds Gracilaria chouae and G.lemaneiformis are widely cultivated seaweed in China.Polysaccharides from seaweeds have biological activity and display excellent biological characteristics.They are ideal material for functional foods and medical materials.In this study,we extracted and separated the polysaccharides from the two seaweeds,and then studied their anti-inflammatory and anti-tumor activities to provide scientific basis for the medical use of seaweeds.The main results are as follows:1.The anti-inflammatory effects of polysaccharides were evaluated with Cell Counting Kit(CCK-8)and Quantitative Real-time-PCR(qPCR).The results showed that G.chouae polysaccharides can significantly reduce the mRNA expression of inflammatory cytokines IL-1?,IL-6,and TNF-? in RAW264.7 and THP-1 cells stimulated by LPS.G.lemaneiformis polysaccharides could significantly reduced the mRNA expression of inflammatory cytokines IL-6,and TNF-? in RAW264.7 and THP-1 cells stimulated by Lipopolysaccharide(LPS).The polysaccharides from G.chouae and G.lemaneiformis exhibit significant anti-inflammatory effects in vitro.2.In vivo anti-inflammatory effects of polysaccharides were evaluated with DSS-induced mice colitis model.The polysaccharides from G.chouae and G.lemaneiformis exhibitd significant anti-inflammatory effects in vivo.The results showed that when G.chouae polysaccharides were administered at a dosage of 250 mg/kg,the disease severity in mice could be relieved.G.lemaneiformis polysaccharides can relieved the severity of disease,the colon was shortening and the colon tissue damage in mice.3.The anti-cancer activity of the two polysaccharides ware evaluated by CCK-8 kit.The polysaccharides from G.chouae could reduce the cell visibility of esophageal cancer cell lines.The experimental results showed that the polysaccharides of G.chouae could reduce the cell viability of esophageal cancer,and especially reduce the cell viability of esophageal cancer ECA109 cell line after they were combined with 5-Fu.However,G.lemaneiformis polysaccharides had no statistically significant anti-esophageal cancer activity.4.The anti-esophageal cancer mechanism for polysaccharides to promote 5-Fu activity was studied with Western blot,and fluorescence microscopy.G.chouae polysaccharides inhibited the autophagy in tumor cells by inhibiting the function of lysosomes,resulting in cell metabolism disorders and subsequent apoptosis.G.chouae polysaccharides promoted ECA109 cell apoptosis by inhibiting autophagy.The polysaccharides of G.chouae and G.lemaneiformis showed anti-inflammatory activity both in vitro and in vivo.G.lemaneiformis polysaccharides can alleviate intestinal inflammation and protect the intestine,which is expected to be developed as an intestine health product or anti-inflammatory medicine in functional food and biomedical fields.Sine G.chouae polysaccharides showed inhibit autophagy of tumor cells and help to solve the problem of chemotherapy drug resistance,it is expected can develope as a chemotherapeutic drug sensitizer,antitumor auxiliary material or functional,medicine.
