Photodynamic therapy(PDT)is a microtrauma treatment for a variety of cancers and certain non-malignant lesions.PDT requires photosensitizer,which can be stimulated by light to produce reactive oxygen species(ROS)with cytotoxicity.This method has been approved for treatment of multiple types of tumors,such as prostate,neck and esophageal cancer,while photosensitizer is indispensable to the photodynamic therapy.However,the commonly used photosensitizers have poor solubility in water,and low optical penetration depth in biological tissues,which restrict their application in PDT.Therefore,we developed two kinds of bifounctional therapeutic agents for cancer diagnosis and high efficient photodynamic theray.Firstly,we designed and prepared the bifounctional water-soluble palladium(II)porphyrin complex for hypoxia imaging and high efficient photodynamic therapy.Secondly,we prepared the Ir-Mn binuclear metal complex for photodynamic therapy.1.Design,synthesis and application of a new kind of the water-soluble Pd(II)porphyrin complex.We developed a new kind of water-soluble Pd(II)porphyrin complex which was formed by quaternizing the oligomeric fluorine that connected with metal palladium porphyrin.The structure of this complex was characterized by using nuclear magnetic resonance(NMR)and mass spectrometry.The physical properties of the Pd(II)porphyrin complex were investaged by spectroscopy,which shows that the complex has favourable stability in aqueous solution.Meanwhile,the palladium porphyrin complex can generate singlet oxygen efficiently under light irradiation and induce cancer cell apoptosis,which can achieve photodynamic therapy.2.Design,synthesis and application of Ir-Mn binuclear metal porphyrin complex.Ir-Mn dinuclear metal porphyrin complex,which formed by connecting iridium complex and manganese porphyrin,has effective photodynamic therapy effects.Then the Ir-Mn dinuclear metal porphyrin complex was encapsulated by DSPE-MPEG5000 to improve its solubility in water.The diameter of the nanoparticles is about 100 nm,which ensure the application of Ir-Mn dinuclear metal porphyrin complex in PDT.The physical properties of Ir-Mn dinuclear metal porphyrin complex were investageted by spectroscopy,and the morphology was characterized by using transmission electron microscopy(TEM)and dynamic light scattering(DLS).Based on those results of the experiments,we concluded that Ir-Mn dinuclear metal porphyrin complex has strong visible absorption,good light stability,pH stability and excellent biocompatibility.All these results suggested that the Ir-Mn dinuclear metal porphyrin complex has a good application prospect in photodynamic therapy.
