Flaxseed is an important vegetable oils resource in Inner Mongolia.Low temperature defatted flaxseed meal,the by products of low temperature oil extraction,which has higher protein content.Flaxseed meal is treated as livestock feed,which leads to a waste of this high quality resource.Therefore,it is of great significance to prepare peptides of flaxseed protein which can increase cholesterol reduction activities for preventing and treating hyperlipidemia.Our project is based on low temperature defatted flaxseed meal,and extracting flaxseed protein.Then enzymatic hydrolysis,ultrafiltration separation is used for peptide preparation which can decrease cholesterol active.In addition,we also discuss the cholesterol reduction function of peptides of flaxseed protein.The main conclusions are:First,as described above,using alkali-solution and acid-isolation method extracting flaxseed protein,and the basic chemical components are determined,the results show that flaxseed protein content of protein,moisture,ash and total sugar is 83.50%,7.93%,1.73% and 6.74% respectively;The analysis of SDS-PAGE gel electrophoresis shows that flaxseed protein have five main subunits,whose molecular weight is 35.70 kDa,24.00 kDa,19.15 kDa,12.26 kDa,and 11.41 kDa.The results also showed that flaxseed protein contains 18 different amino acids.Therefore,the flaxseed protein extracted in this experiment is of high purity and rich in nutrition.And then,when we use Protease M as our digestive enzyme to prepare hypocholesterolemic peptides and detect different digestion time products,the results show that when the digestion time for 4h,flaxseed protein hydrolysatesc has the highest cholesterol micellar solubility inhibition rate,57.98%.The binding rate on cholic acid sodium,sodium taurocholic acid,and sodium deoxycholic acid was 44.39%,31.17% and 57.44%,respectively.Further,the ultrafiltration technology was used to the flaxseed protein hydrolysates.The result shows that when the molecular weight ?1kDa,the contents have highest cholesterol reduction activities.The cholesterol micellar solubility inhibition rate was 72.39%,the binding rate on bile acid sodium,sodium taurocholic acid,sodium deoxycholic acid was 63.18%,49.80% and 67.20%,respectively.In this paper,the lipid-lowering effect on hyperlipidemia rats was investigated by ultrafiltration separation of flaxseed protein hydrolysates with hypocholesterolemic which molecular weight ?1kDa.The result shows hypocholesterolemic peptide derived from flaxseed protein can reduce body weight of rats,reduce the AI,and positively affect abnormal blood lipid of hyperlipidemia rats.The hardening of the arteries was reduced in high fat rats with increasing levels of high density lipoprotein-cholesterol(HDL-C)in serum.Proteolytic peptides of flaxseed have certain positive effects on rat spleen and can also repair fatty degeneration of the liver cells.
