In this thesis,thermosensitive monolayers were prepared using?3-mercaptopropyl?-trimethoxysilane and?3-methacryloyloxypropyl?trimethoxysilane functional groups,respectively.The experiment was designed using spass19.0 software.The optimal conditions for preparation are as follows:the mass of monomer NIPAM is 0.4500g?30%?,initiator concentration is 20mg·mL-1,reaction temperature is 110°C,heating time is 2.5h.The two temperature-sensitive monolayers adsorbed myoglobin and bovine serum albumin respectively.The UV spectra showed that the?3-methacryloxypropyl?trimethoxysilane monolayer membrane had wider reversible adsorption protein range;In order to apply temperature-sensitive controlled protein adsorption to capillary,pyrolysis method,physical method and photolysis method were utilized to overcome shortcomings of traditional capillary monolithic column.This paper provides a certain reference for the protein separation and enrichment in the capillary.
