A great deal of discarded persimmon peel is produced in the persimmon process,the persimmon peel contains rich functional nutrients.In order to improve the value of persimmon peel,the extraction method of persimmon peel pigment were explored by ultrasonic solvent extraction,the composition of persimmon peel pigment is analyzed by high pressure liquid chromatography,the chemical stability,antioxidant activity,bacteriostatic,anticancer functions in vitro of persimmon peel pigment was investigate.The results show as follow:?1?The crushed dried persimmon peel was extracted for 2h with 10 times 95%ethanol at50?under pH7 by 3 times ultrasonic extraction,the pigment extraction effect was best.The pigment extract was washed and precipitated for several times with 10 times water after vacuum rotary evaporation recovery.The pigment was collected and dryed under vacuum,the yield of persimmon pigments was up to 21.8g/kg.?2?The persimmon peel pigment contains at least 10 components by HPLC analysis,which are mainly 52.31%?-carotene,8.73%lutein,and 4.02%zeaxanthin.The IC50 values of persimmon pigments on DPPH,ˇOH,ABTS+ˇfree radical Scavenging ability and Fe2+chelating ability were 0.44,0.27,0.37 and 0.35mg/mL respectively.Compared with Vc,?-carotene and lutein,the antioxidant properties were Vc>beta carotene>persimmons peel pigment>lutein,and the persimmon peel pigment resisting lipid peroxidation ability is raised with increasing concentration,proving that the pigment of persimmon peel had certain antioxidant capacity in vitro.?4?The persimmon peel pigment would severe fading under illumination,high temperature,in oxidation agents and Cu2+solution,producing precipitation in acid,Ca2+,Zn2+,Al3+solution,while the reducing agent and preservatives have protective effect on the persimmon peel pigment.?5?The minimal inhibitory concentrations?MIC?and the minimum bactericidal concentration?MBC?of the persimmon peel pigment on the three kinds of pathogenic bacteria?E.coli.,S.aureus and Salmonella?is 10,5,5 mg/m L and 20,10,10 mg/mL,respectively.However the persimmon peel pigment has not inhibitory effect on Listeria and Ssakazakii.The proliferation inhibition effect of persimmon peel pigment on human liver cancer HepG2 cells and gastric cancer MNK-45 cells are not obvious.
