| China has a wide variety of acorn resources.Acorns are rich in plant polyphenols and have biological activities such as anti-aging,anti-cancer,antibacterial and anti-inflammatory activities.In order to develop acorn polyphenol functional health products.This article used Quercus Variabilis acorn nutlets as the experimental material,by solvent extraction,macroporous resin adsorption and purification,UV spectroscopy,infrared spectroscopy analysis,and HPLC analysis,to study the composition of polyphenols in Quercus Variabilis acorn nutlets,combined with the vitro antioxidant activity,antibacterial activity functional evaluation was performed.The main results were shown as follows:1.Acorn nutlets powder were ultrasound assisted extraction with ethanol.The crude polyphenols extract ratio of acorn nutlets was 20.5%,and the content of gallic acid in rude polyphenols was 345 mg/g.The D4020 macroporous resin is the most suitable resin for the purification of polyphenols from acorn nutlets.After purified from the D4020 macroporous resin,the content of gallic acid in crude polyphenols was 702 mg/g,and the purity was increased by a factor of one.Ultraviolet absorption spectrum analysis of acorn nutlet pure polyphenols showed that there were two characteristic absorption peaks at 270 and 365 nm,which were in line with the absorption characteristics of polyphenols.The results of infrared absorption spectra showed that acorn nutlet pure polyphenols had characteristic absorption at3646,1732,1616,1455,1354,1190,1055cm-1.The structure of acorn nutlet pure polyphenols contains hydroxyl,carbonyl,aromatic ring(C=C)and ortho-substituted aromatic(C-H)structures.The HPLC analysis showed that the acorn nutlet pure polyphenols contained at least six polyphenolic substances such as protocatechuic acid,caffeic acid,ferulic acid,ellagic acid,quercetin,and kaempferol.The highest content of caffeic acid in acorn nutlet pure polyphenols was 24.9 mg/g,followed by protocatechuic acid 14.6 mg/g,ellagic acid 3.2 mg/g,ferulic acid 2.2 mg/g,and less quercetin and kaempferol content.2.The DPPH radical scavenging activity of acorn nutlet crude polyphenols and pure polyphenols was pure polyphenols>Vc>crude polyphenols;The ABTS radical scavenging activity of acorn nutlet crude polyphenols and pure polyphenols was Vc>pure polyphenols>crude polyphenols;the total reducing power of crude polyphenols and pure polyphenols was not as good as that of Vc;and the metal iron chelating abiliyt of acorn nutlet polyphenols was not as good as that of EDTA-2Na;The anti-lipid peroxidation ability was not as good as that of Vc.The acorn polyphenols have a certain antioxidant capacity,but the effect is not strong.3.Acorn nutlet crude polyphenols and pure polyphenols had significant inhibitory effects on platelet,Escherichia coli,Staphylococcus aureus,and Salmonella.The effect of pure polyphenols was superior to that of crude polyphenols,The antibacterial activity of pure polyphenols was similar to that of potassium sorbate.Using acorn nutlet pure polyphenols to treat bacteria,its growth rate was significantly slowed down.The acorn polyphenols have a strong antibacterial function. |
