It is very important to develop new and simple biological functional materials,and study its application in biochemical analysis and clinical treatment.In this paper,the preparation of new conjugated oligomer nanoparticles and its applications in cells target imaging,photodynamic therapy and drug-loaded therapy were studied.The main research contents are as follows:1.The preparation of oligonucleotide(DNA)-functionalized conjugated oligomer nanoparticle(CON)and its target imaging of cancer cells.First,we used reprecipitation method to prepare new type of CON.Through EDC/NHS reaction,we linked the folate modified single-stranded(DNA)to the surface of CON for directly forming functionalized CON.Then,the functionalized CON was incubated with A549 cells,and specifically bind to the surface of A549 cells that have the folate receptors.The preparation method of CON is sample,low cost,and the CON has a high fluorescence quantum yield,good biological compatibility,which provides a new nanomaterial for targeted fluorescence imaging of cells.2.The photodynamic and drug-load therapy of cancer cells by the functionalized CON of was developed.Under the exposure of xenon lamp,CON can produce reactive oxygen species(ROS),which can further induce cancer cells death.Thus,when CON targeted to the cells,CON can be used to kill cancer cells by photodynamic therapy.Furthermore,a single-stranded DNA was designed to hybridize with the folate-modified DNA and to form double-stranded DNA.Subsequently,the doxorubicin(Dox)was embedded in the GC base pairs of the double-stranded DNA.Then,we incubated the Dox-loaded CONs with cancer cells.The CONs would enter the cells through Cell endocytosis,leading to the release of Dox to kill cells.And we further demonstrated that the concentration of CON can be reduced by the combination of photodynamic and drug therapy of CONs for cell killing.The multifunctional CONs provide a new method for cell killing based on the photodynamic and drug therapy.
