The dissertation consists of two independent parts.Part?Simultaneous determination of mannitol and five sugars in urine by gas chromatographyA simultaneous determination of mannitol,lactulose,sucrose,frutose and glucose method was established by gas chromatography,using inositol as the internal standard.The samples were handled with oximation and silylation before chromatographic analysis.All compounds were obtained good separation under the optimal conditions.The linearity of mannitol,lactulose,sucrose,fructose and glucose was good and the linear coefficient was more than 0.999;The RSD of inter-day precision was less than 2.69%;the RSD of intra-day precision was less than 3.90%and the recovery of three different concentrations ranged from 85.13%to 109.78%,which indicated that the method was simple,senstive,accurate and could satisfy clinical need.Part?The detection of free fatty acids in serum based on serially coupled columns in gas chromatographyFree fatty acids were one of significantly important active substances in our body.The transformation of their compositions,the change in concentration and the metabolic biotransformation could reveal pathogenesis and development of diseases,moreover,they could evaluate therapeutic effects and deeply study on the mechanism of medicines.In this study,a convenient and stable method of 0.4 mol/L KOH-CH3OH and 10%H2SO4-CH3OH was used to extract and derive free fatty acids in serum respectively,then different combination types of column were obtained through research.The results showed that SLB-IL-111 as 1D and TR-5MS as 2D were the optimal combination.Next,methodology validation of free fatty acids in serum based on serially coupled columns gas chromatography was established.The linear coefficient was more than 0.999;The recovery of free fatty acids of three different concentrations ranged from 83.33%to 110.09%;The RSD of inter-day precision was less than 5.48%;the RSD of intra-day precision was less than 6.31%.The method was of high speed,accuracy and high throughput and could satisfy the need of clinic,and which would provide good reference for researching the relationship between metabolism of free fatty acids and diseases or therapeutic effects.
