The purification technology of Chinese medicine is one of the bases for ensuring the safety,effectiveness and hygiene of Chinese herbal medicine preparations.The modernization of traditional Chinese medicine has put forward higher requirements for the traditional technology based on alcohol precipitation,and the subsequent flocculation technology has been the hope of the industry.In the case of poor effect of single flocculation on the removal of impurities in traditional Chinese medicine,this paper explores the purification of XiaoYanTuiRe(XYTR)raw water extracts by the combination of chitosan and chitosan quaternary ammonium salt(HACC),and analyzes the synergistic effect of the compound flocculant and the selective expression mechanism of the flocculation technology.In this paper,the retention rate of the effective component of acetotin(RT)and the removal rate of impurity protein(RM)in the purification process of XYTR water extracts and the turbidity of the liquid supernatant after flocculation(ST)are used to determine the condition selection of the flocculation process.On the basis of orthogonal experiment,the effect of flocculant dosage(CM),flocculation temperature(TO),impeller line velocity(SF)and stirring time(tf)on flocculation effect of chitosan and HACC were studied.The selective expression mechanism of flocculation technology and the observation and judgement of the floc strength formed by different flocculation mechanism were studied by comparison and analysis of the particle size distribution map and micrograph of the flocculating liquid.The selective expression of flocculation mechanism presents a cross grain size subdivision phenomenon in the size distribution of the liquid.The synergistic effect of chitosan and chitosan quaternary ammonium salt is that chitosan catches small particle size particles,chitosan quaternary ammonium salt catches large particle size particles,and the size of the solution is D50=3.738 m under the compound ratio of 2:1,and the micromorphology of the flocculated liquid is small powder,uniform single,and all indexes are good.The regression equation of the retention and impurity removal rates of liquid medicine was established by using MINTAB software,and the simplified model was determined by analysis of variance(ANOVA)and residual error analysis.The analysis of the equivalent line and response surface of the simplified model was carried out,and the significant level of the experimental factors and their two order interaction effects was obtained with the response variables of the retention and impurity removal rates of the liquid.Finally,the optimization process of compound desirability D=0.7303 is obtained by response optimizer.The optimization results are CM=1.150,TO=50,SF=500r/min,tf=2min.Under this condition,RT=87.60%,RM=88.29%.The combined application of chitosan and chitosan quaternary ammonium salt in eliminating inflammation and antipyretic Chinese medicine water extract has reference significance for actual production.
