Droplet-based microfluidics uses immiscible oil to form picoliter to nanoliter-scale droplets,which reduces sample dilution and loss effectively.Meanwhile,the dropelts enclosed with immiscible oil can eliminate sample absorption on the solid channel surface and provide biocompatible environments.Recently,droplet-based microfluidics has been applied in biology,medicine,and pharmacology.A two-dimensional droplet array spreads the droplets on the plane of a chip,and the precise positioning of each droplet can be achieved by the two-dimensional coordinates of the droplets,which facilitates long-term observation,selection and subsequent study of the target droplets.In charpter 1,the methods used to form two-dimensional droplet array were reviewed.According to the system design,the methods were divided into two major categories:generating droplet arrays in closed systems and generating droplet arrays in semi-open systems.The chip structure and the process of forming droplet array were further introduced.In charpter 2,we developed the method to generate oil-in-water droplets and water-in-oil-in-water droplets,a chip to form high-throughput two-dimensional droplet array in seconds,and a platform to pick active droplet and culture the target cell.We first established a polydimethylsiloxane cross-channel chip to generate primary and secondary droplet.We optimized the chip structure and droplet spreading methods to form droplet array.We combined the droplet array with fluorescent detection to screen active droplets and culture the single cell in the droplets.We applied this method to the generation of a million-scale two-dimensional droplet array in several tens of seconds and the selection and cultivation of active single cell droplets.
