The enhanced biological phosphorus removal process is an efficient and stable sewage treatment process developed in the practice of long-term water treatment.The process has been widely used in the control of water pollution,and significant progress has been made in the research of its treatment mechanism.Many new processes have also been derived on it.However,as a representative microorganism,“polyphosphate bacteria”(general name for phosphorus removal microorganisms,PAOs),the metabolic details are still unclear.So far,no pure PAOs have been isolated,which leads us to lack the tools for quantifying them.This has hindered the further understanding of the phosphorus removal mechanism and subsequent research.By operating and maintaining the enhanced biological phosphorus removal reactor,the optimal debugging interval of each impact factor was determined,and the intracellular polyphosphate was quantitatively evaluated by Raman microscopy,in order to quickly characterize its metabolic details and elucidate the intracellular state of the microbial population.Relationship with system performance.This topic mainly studies the following aspects:1.The operating and maintaining of the enhanced biological phosphorus removal reactor: This project selects the return sludge of the wastewater treatment plant for inoculation,and carries out the smoldering treatment to enhance the activity.The sodium chloride is constructed by using the sequencing batch reactor(SBR).The biological phosphorus removal system,which is the sole carbon source,is cultured by periodic cycle of anaerobic stirring,aerobic aeration,sedimentation and periodic drainage,in order to achieve the screening of microorganisms in the sludge,so that the polyphosphate bacteria become the dominant bacteria.For the purpose of this,during the period,the operating parameters such as MLSS,SVI,DO,etc.are regularly checked to maintain the long-term operation of the reactor.2.Determination of the optimal debugging interval of each impact factor: change the factors such as p H,dissolved oxygen,C/P and sludge concentration,determine the optimal debugging interval through the operation effect of the reactor,and ensure the long-term stable operation of the whole system..It is confirmed that the dissolved oxygen in the anaerobic section should be maintained below 0.24 mg/L,and the dissolved oxygen in the aerobic section should be maintained between 2 and 4 mg/L;and the optimum range of p H is 7to 7.5;P should be maintained between 10 and 15.3.Characterization of enhanced bio-phosphorus removal by micro-Raman: By detecting the Raman spectrum of freeze-dried sludge and smoothing and normalizing it after Savitzky-Golay,Gaussian fitting is combined with partial least two Multiply(PLS)establishes a model between spectral data and chemical data,and the correlation between the two models reaches 0.8081.The above indicates that microscopic Raman characterization can qualitatively and quantitatively characterize PAOs.
