Inonotus obliquus is a very rare and valuable edible multi-functional fungus.According to experiments,it has many biological activities,such as anti-tumor,AIDS prevention,immunity enhancement,diabetes prevention,anti-oxidation,lowering blood sugar,blood lipids and other effects.In this paper,the with polysaccharides of Inonotus obliquus as the research object,through the study of different doses of polysaccharides of Inonotus obliquus on immunosuppressive model mice immune activity,the does-effect relationship of polysaccharides from Inonotus obliquus were determined.Through the research on the influencing factors of the immunological activity of polysaccharides of Inonotus obliquus,the influence of processing conditions such as temperature,pH,and irradiation on the immune activity of the products was established,thus laying the foundation for the product development and comprehensive utilization of Inonotus obliquus.According to the results of the quantitative effect study,combined with the optimal dose at the age in days,single-factor tests,orthogonal tests,and response surface analysis methods were used to optimize the best product formula for the immune yoghurt of Inonotus obliquus and Inonotus obliquus.We developed a functional product of Inonotus obliquus,which is known to enhance human immunity activity,inoculum inoculum and immune cells of Inonotus obliquus.Research indicates:(1)According to the results of immune activity studies,the mice were selected to be injected intraperitoneally with cyclophosphamide for 3 consecutive days to establish an immune activity model of the test mice,and the mice were each intragastrically administered 50 mg/kg,100mg/kg,200mg/kg,400mg/kg,800mg/kg for 15 days of the polysaccharide of Inonotus obliquus.Based on the change in weight gain of the immune activitied mice during the test,When the dose of the polysaccharide of Inonotus was between 50mg/kg and 100mg/kg,the weight of the activitied mice increased from 3.90 g to 4.04 g.and the increase in body weight of immune activitied mice was greatest at polysaccharide doses between 200mg/kg and 800mg/kg garound;by measuring the mouse organ index,when the polysaccharide dose was between 50mg/kg-200mg/kg,immune suppression mouse thymus index increased from 3.217mg/kg to 3.773mg/kg,spleen index increased from 3.477 mg /kg to 3.700mg/kg,when the Inonotus obliquus polysaccharide dose 400mg/kg-800mg/kg,immune activitied mice thymus index and spleen index were the largest,respectively about 4.2mg/kg,and 3.8mg/kg,with the best repair effect on injured immune organs;the results of the effect on humoral immune function were as follows: when the dose of polysaccharide of Inonotus obliquus was between 50mg/kg-200mg/kg,the OD value of the serum hemolysin antibody was increased from 0.292 to 0.351,and when the polysaccharide was between 400mg/kg-800mg/kg,the maximum of DVvalue is about 0.37,and the level of hemolysin antibody in the mouse serum is the best;through the effect analysis on the function of mononuclear-macrophage,the dose of polysaccharide of Inonotus obliquus is between 50mg/kg-100mg/kg.The phagocytic index ? also increased correspondingly from 3.752 to 4.138.When the polysaccharide dose was between 200 mg/kg and 800 mg/kg,the phagocytic index ? was maximal about 5.7,and the phagocytic function of the mouse macrophages was the strongest.Therefore,in summary,the dose-effect relationship for the polysaccharide immune activity of Inonotus obliquus is found to be the best when the polysaccharide content is between 400 mg/kg and 800 mg/kg.(2)The results of the influencing factors of the polysaccharide activity of Inonotus obliquus polysaccharides showed that the polysaccharides treated with sterilization conditions of 80°C,100°C,and 121°C,and their weight,thymus and spleen index,serum Hemolysin antibody and mononuclear-macrophage phagocytosis index had no effect;pH 4.0,pH 5.0,pH 6.0 and other pH conditions of the treatment of the polysaccharide,it has no effect on the immune activity indicators of mice;electron beam radiation The polysaccharides treated with various irradiation intensity conditions such as 1kGy,3kGy,and 5kGy had no effect on the immune activity of the mice;the polysaccharides treated under various irradiation conditions such as 2min,4min,and 6min of ultraviolet irradiation were tested.The immune activity of mice had no effect;polysaccharides with ionic strengths of 0.4 mol/L,0.6 mol/L,0.8 mol/L,and other ionic strength conditions had no effect on the immune activity of mice,indicating that the product processing technology and processing Conditions will not affect the increase of human immune activity against Inonotus obliquus.(3)According to the results of dose-response relationship research,the single-factor test and orthogonal test were used to evaluate the yoghurt sensory score.Poria polysaccharide 0.7%,white sugar 7%,pectin 0.2%.At this time the yoghurt is yellow-brown and has the inherent flavor of yoghurt and the proper bacillus of birch brown.The surface is smooth,the texture is fine and uniform,there is no precipitate output,no whey and other substances precipitate,and the developed product meets the product quality standard.(4)According to the results of dose-response relationship,using single-factor test and response surface test,the sensory score of cheese was used as the evaluation index,and the product formula of the immune cheese of Inonotus obliquus was selected as follows: Inonotus obliquus polysaccharide 0.8%,starter 1.03%,rennet enzyme 1.27%,fermentation time 6 days.At this time,the color of the cheese is uniform,glossy,yellowish-brown,with milk flavor and appropriate flavor of Inonotus obliquus,and the cheese is stable,soft and delicate,moderate in viscosity and hardness,easy to cut,and the developed product meets product quality standards.
