Chitosan and collagen have good biocompatibility and biodegradability and are important biomedical materials,which play an important role in the field of tissue repair.In this study,the main factors affecting biological sponge dressings were explored by using chitosan and collagen as the main materials with the method of orthogonal experimental to evaluating evaluate the physical properties of sponge dressings under different conditions.It was shown that when the total mass fraction was 2.5%,its optimal preparation process was as follows:the ratio of chitosan to collagen was 2:1;the concentration of crosslinking agent glutaraldehyde was 0.015%;the crosslinking temperature was 4?and the crosslinking time was 4 hours.The sponge dressing prepared by this process had good water absorption and breathability and lower process cost.The biological safety was detected by animal experiments.It was shown that the self-made sponge dressing had good biocompatibility.In this study,the effect of sponge dressing on wound healing was studied by using mice skin incision model.It was shown that the optimal healing was sponge with FGF1 group.HE staining and Masson staining were used to observe the wound regeneration of mice with different dressings.It was found that using sponge with FGF1 groups~'wound regeneration was more complete and collagen was higher,which proved that sponge with FGF1 could promote collagen deposition to promote wound healing.The expression of inflammatory response-related genes and apoptosis-related genes in mice wound tissues were detected by real-time fluorescence quantitative PCR.The results showed that sponge dressing with FGF1could inhibit the expression of IL-1?,TNF-?and IL-18 to reduce the local inflammation of the wound to promote wound healing and improve the quality of wound healing by eliminating the extra inflammatory cells by promoting the expression of apoptosis-related genes Caspase-3 and Bax.
