Folic acid is a pteroic acid mono glutamate.It is one of the vitamin B complex,which is used as anti-anemia medicine used during pregnancy and infantile megaloblastic anemia embolism treatment.Now,the need of folic acid in China increases largely every year.Therefore,it is very important to produce it in industry from both points of medicine and economics.However,the existing process for the manufacture of folic acid has the disadvantage of generating large amount waste water causing serious environmental pollution,therefore,it is limited that folic acid is manufactured in large scale.Therefore,the development of green process for the production of folic acid is necessary.First,we optimized the traditional production process of folic acid.The reaction of L-N-aminobenzoyl glutamic acid,2,4,5-triamino-6-hydroxypyrimidine sulfate and 1,1,3-trichloroacetone was run in 50% aqueous solution of ethanol instead of pure water to synthesize folic acid,which reduced the water consumposition of 93%.After the reaction,the crude product was purified by the process of solubilization in acid,precipitation by addition of water,re-solubilization in alkaline,decolorization with active carbon and precipitation with acid to afford folic acid.The effects of reaction conditions on the yield and purity of folic acid were investigated and the the optimum conditions were obtained.Under the optimized reaction conditions the yield of folic acid was increased obviously.It was found that the crystalline 1,1,3-thichloro-acetone can be obtained from the recrystalization of crude 1,1,3-thichloro-acetone from petroleum ether–water mixture.The yield of folic acid was increased about 10% by using crystalline 1,1,3-thichloro-acetone as raw material to react with L-N-aminobenzoyl glutamic acid,2,4,5-triamino-6-hydroxypyrimidine sulfate.Then,the new method to synthesize folic acid was investigated.?-Bromopyruvaldoxime,?-chloropyruvaldoxime and 2,2,3,3-teramethoxypropanol were synthesized to repalace 1,1,3-trichloroacetone as the intermediates respectively in the synthesis of folic acid.Though the attempts were failed,the reasons for the failure were screened.
