| With the extensive application of nuclear energy,the effect of ionizing radiation on the health of human body has attracted more and more attention.Copper and iron in organisms are closely related to many kinds of physiological activities.Studies show that copper and iron mainly exist in two kinds of chemical forms including free form and combined form,metal ion in free form can produce biological toxicity,while combined metal ion is in favour of producing beneficial biological effect,so that metal ion in different chemical form has extremely different biological effect.In this study,concentration of copper and iron in different chemical forms and changes of valences states in irradiated mice serum was detected by flame atomic absorption spectrophotometry and chemical colorimetry respectively,aiming to detecting the effect of irradiation on copper and iron in different chemical form.The research mainly has three aspects as following:1.Study of copper variation in different chemical forms under different gamma ray irradiation conditions The result showed that concentration of uncombined copper in irradiated mice serum had no significant change,the concentration change trend of combined copper in mice serum with different dose gamma irradiation rise which was caused by concentration change of ceruloplasmin,the peak point changed with irradiation dose increased;Moreover,the linear relationship for copper concentration of the dose-effect relationship was significant at 24 h after irradiation in mice serum.2.At the same time,the study for iron was ongoing.The result showed that concentration change of iron was mainly besed on the combined iron which declined after irradiation,and the decline was caused by ability enhancement of cell absorbing iron in serum with concentrations of transferrin in serum and transferrin receptor rose after irradiation.Concentration linear relationship of the dose-effect relationship was significant at 10 h after irradiation in mice serum.3.Study of relevance between combined copper and iron.The result of experiment showed that with concentration and activity of ceruloplasmin in serum of irradiated mouse rose,Fe2+was oxidized into Fe3+,meanwhile as the active center of ceruloplasmin,Cu2+was reduced into Cu+.For further proof about the relevance,mice got pretreatment with GSH to inhibiting free radicals.The concentration of Fe2+in treated mice was higher than untreated mice after irradiation,but much lower than normal mice,proved that decline of Fe2+was caused by ceruloplasmin beside free radicals.In the study,changes of copper and iron in different chemical under the condition of irradiation was detected,experiment results provide valuable reference for further study in biological effect of irradiation about copper and iron. |
