In our study,we serve Jilin Province specialty blueberries as the main raw material,and other traditional Chinese medicine as compound materials including Gardenia,Turmeric,Salvia,Centella,White peony.The Blueberry Jiuganqing capsules?BJC?were made up by raw materials of extraction and preparation processes.After that,we examined BJC exerts a protective effect against alcohol-induced hepatic injury,and the result can apply for the health food symbol.Our study made a detailed study on the preparation technology,the quality standard of the finished product,the preliminary stability,protection of hepatotoxicity and acute toxicity of BJC,this provides experimental basis for further development and application of BJC.The extraction and preparation processes of BJC:Blueberry,Gardenia,Turmeric and Salvia were mixed according to the prescription ratio,and the active ingredients were extracted using ethanol reflux extraction method.The content of the geniposide and the extraction rate were determined using the L9?34?orthogonal test.The data were analyzed by comprehensive score method,the best extraction process was 70%ethanol,the liquid-solid ratio was 1:15,refluxed for 2 times and 3 hours everytime,filtered,and the filtrate was combined and concentrated into a dry paste,recovered ethanol;The active ingredients of Centella and White peony were extracted by water decoction,and determined by L9?34?orthogonal design method.The content of paeoniflorin and extraction rate were as the investigated index,the data were analyzed by comprehensive score method,the best water extraction process was solid-liquid ratio 1:15,water extracted for 3 times and 3 hours every time.After repeated experiments,the best forming process of the capsules were that the soluble starch and the dry extract powder were mixed in 1:1 mass ratio.The obtained powder had good fluidity and less hygroscopicity.The study on quality standard of BJC:Thin layer chromatography?TLC?was used to qualitatively identify the components of Centella,Turmeric,and Salvia,the spots are clear,the method is simple and accurate,and the negatives have no interference.HPLC method was used to determine the content of geniposide,paeoniflorin and UV spectrophotometry was used to determine the content of total flavonoids.The method was simple,accurate,stable and reproducible.BJC routine examination:moisture,loading differences,disintegration time are in line with the provisions of Pharmacopoeia.The experimental study on the preliminary stability of BJC:A three-month accelerated test and a six-month long-term stability test were performed on three batches of samples,respectively.No significant change in traits.In the TLC identification items,Centella,Turmeric and Salvia can be detected.BJC Routine Examination Meets Pharmacopoeia Requirements.There was no significant change in the content of the indicator components.The results show that the product is relatively stable.The study on the protective effect of liver function of BJC:Totally 60 mice were randomly divided into control group,model group,BJC-L?150mg/kg?,BJC-M?300mg/kg?,BJC-H?600mg/kg?and positive control group?60mg/kg?.Mice were pretreated with BJC by oral gavage for 30 days before being treated with 12 ml·kg-1alcohol,severe liver injury was exerted,and hepatotoxicity was assessed.Our study results demonstrated treatment with BJC significantly decreased the serum alanine aminotransferase?ALT?,aspartate transaminase?AST?,triglyceride?TG?levels compared to the alcohol administration.Meanwhile,compared with that in the alcohol group,PQS decreased hepatic malondialdehyde?MDA?contents and restored reduced glutathione?GSH?content and superoxide dismutase?SOD?activity in livers of mice in a dose-dependent manner.The present study clearly shows that BJC exerts a protective effect against alcohol-induced hepatic injury.The experimental study on safety of BJC:40 mice were randomly divided into two groups:the treatment group and the normal group.Mice were pretreated with BCJ?0.4mL/10g?by oral gavage for 14 days,and the normal group was given an equal volume of distilled water,dosed three times within 24 hours.The results showed that,daily doses were administered at a maximum dose of 18g/kg and no acute toxicity reaction in mice,no abnormalities after organ dissection,and compared with the normal group,the mice body weight,food intake,and organ coefficient were not statistically significant.The BJC is a low-toxic and safe health product,which provides a basis for further clinical research.
