Hyaluronic acid(HA)is a mucopolysaccharide with unique molecular structure and the physical and chemical properties.It has been successfully used in cosmetics,arthritis therapy and medicine.The research was mainly to study the extraction and purification process of hyaluronic acid from marine resources—tuna to get optimum technological conditions.First of all,thawed out tuna eye and get the vitreous,use acetone degrease,then crush,and decompression drying,get powder.Then optimize the extracting conditions.The results show that,when the material liquid than 1:15,at the temperature about 20 ?,extracting for 4 hours,could obtain 2.46 mg HA per dry powder.Secondly,the combination of enzymic digestion and alcohol precipitation were carried out for the protein elimination of extracting solution.The final conditions: 0.2% of papain,pH 6,60 ?,reaction for 2hours,then add triple 95% ethanol solution which containing 1%potassium acetate,precipitated 6 h,the protein removal rate get 97.96%.Meanwhile,design the enzyme membrane reactor,and applied to study the removal rate of protein in hyaluronic acid preparation.Theresults show that when the MWCO of ultrafiltration membrane is 10000,at the temperature of 50?—60?,and the pressure of 0.15 MPa,the retention rate of HA is 60.65%,with the protein removal rate of 86.34%.Further purification process is Cetylpyridinium chloride(CPC)precipitation,and then by ethanol precipitation,dehydration,drying and other steps,obtaining purified HA.After the CPC precipitation,HA recovery rate of 90.42%.Hyaluronic acid Purity in the final product was 78.36%,protein content 1.48%.This study obtained a simple process route and access to high quality HA products,which lay a foundation for industrial application.
