| Coumarins is a kind of very important organic compounds,distributing in many plants and spices,also presenting in microbes and animal physiological metabolites.Coumarin compounds can be used as fluorescent molecular probe,antioxidant,antibacterial,antivirus,anticoagulant,insecticide laser pigment etc.Because of their natural bioactivities,coumarin derivatives has been widely applied in medical,pharmaceutical,life science,chemical industry etc.fields.Therefore coumarins become an enduring topic in scientific research.In this study,a number of coumarin derivatives is synthesized and subjected to the screening for nematicidal activity against pine wood nematode(Bursaphelenchus xylophilus).Some of coumarin derivatives were reported for the first time,this enriched coumarin derivatives types and expanded its application scope.The first chapter mainly introduces coumarin derivatives classification,application and chemical preparation method.This chapter classify coumarin derivatives by its biosynthesis pathway,they can be classified into simple coumarins,furanocoumarins,Pyranocoumarins,isocoumarin etc.coumarin derivatives fluorescence characteristics applications,antibiotic and antioxidant applications,medical applications,biological science,nematicidal activity and other field applications were reviewed;Coumarin derivative may be synthesized though various ways,such as Perkin,Wittig,Knoevenagel condensation,Baylis Hillman,and Pechmann reaction etc.Pechmann reaction was chosen in this study.Mannich reaction is also briefly introduced;current domestic research status of pine wilt diseases were analyzed and summarized;the last part of this chapter presents the main task of the research and innovation point.The second chapter descripts the synthesis of some basic coumarin skeleton,then further derivatization by Mannich reaction.As the reaction proceeds under mild conditions,with simply operation,it’s facile to get this kind chemical derivatives All the synthesized chemical product structure were confirmed by ~1H-NMR,IR spectrum,and the melting point.3-Methyl-4-isopropyl phenol coumarin,tert butyl hydroquinone coumarin,7-hydroxy-4-methyl coumarin aliphatic amine Mannich bases etc.are new chemical entities,Reports for their synthesis were the first time.The third chapter descripts screening of coumarin derivatives for nematicidal activity against Bursaphelenchus xylophilus.Firstly,pine wood nematode were collected by Biermann funnel method,then was cultured and amplified in the tomato Botrytis cinerca of inoculation on the PDA plate.Isolation amplified of pine wood nematode,continuing amplification under 25℃thermostatic,separating and preparation nematodes tested suspension..The pine nematode mortality was judged by using acid-base indicator neutral red staining method,activity of coumarin derivatives against Bursaphelenchus xylophilus was finally determined.The fourth chapter descripts mainly the discussion of the experimental results.Activity detection results showed that skeleton coumarin derivatives bearing poor solubility,resistance to pine wood nematode activity are also low;Derivatization by Mannich reaction,it’s solubility are greatly improved,but resistance to pine wood nematode activity does not necessarily improved;Retain free phenolic hydroxyl coumarin Mannich bases resistance to pine wood nematode activity are higher than the non-free phenolic hydroxyl coumarin Mannich bases,containing free phenolic hydroxyl coumarin Mannich bases of resistance to pine wood nematode activity are higher than that of Psoralen corylifolia element of resistance to pine wood nematode activity,it’s anti pine wood nematode activity equal to Terpineol or slightly higher. |
