Nanomaterials have inspired great attention in some fields including diagnosis,energy and environment,due to their excellent biocompatibility,unique physical and chemical properties,etc.In addition,the aptamer has been developed as a new recognition molecule and possesses the potent ability of high specificity and effective treatment.So the aptamer functionalized nanomaterials emerge immediately as potential tools in the apoptosis of tumor cells and treatment of cancer.They can induce the apoptosis of tumor cells,and thus exhibits excellent therapeutic effect.Therefore,a variety of therapies have been developed,such as chemotherapy,photothermal therapy,photodynamic therapy,and double or even triple synergistic therapy based on these types of therapies.These scientific researches provide many meaningful results for clinical studies,which are expected to improve tumor cure rate.Based on the biotechnologies and nanotechnologies mentioned,this paper presents an exploration of the use of aptamer functionalized nanomaterials in the apoptosis of tumor cells and treatment.The main results were summarized as follows:1.A Multifunctional Core-Shell Nanoplatform for Enhanced Cancer Cell Apoptosis and Targeted ChemotherapyA novel multifunctional nanoplatform was designed based on the combination of silver nanoparticles with nucleolin-targeted and doxorubicin-loaded manganese dioxide nanosheets to induce enhanced tumor cells apoptosis.MnO2 nanosheets formed on the surface of AgNPs served as efficient fluorescence quenchers of DOX.After internalized into tumor cells,the fluorescence of DOX could be turned on gradually by intracellular glutathione which reduced MnO2 into Mn2+ to release DOX.The synergetic effects of AgNPs-induced apoptosis and subsequent DOX delivery resulted in enhanced tumor cells apoptosis.Annexin V-FITC/PI double staining,mitochondrial membrane potential detection and reactive oxygen species detection demonstrated the specific enhanced apoptosis of tumor cells.In this way,the novel nanoprobes can be used as promising theragnostic agents for specific tumor therapy.2.Aptamer Functionalized Porous Gold Nanostucture for pH?Temperature and NIR-Activatable Targeted Tumor Cell Synergistic TheranosticsA multi-stimuli responsive drug vehicle based on dual-controllable copolymer-liposome bilayer(PLB)capped porous gold nanosponge(GNS)was developed for targeted theranostics.The fabricated gold nanosponge possessed the capabilities of high drug loading.Drug release was controlled by the pH and temperature sensitive PLB,as well as by the NIR irradiation to gold nanosponge.As compared to either of two therapies independently,a considerable inhibition of tumor cells treated with the combination of chemotherapy and photothermal therapy was observed in vivo.Besides,the targeted nanoprobes minimized the harm to normal cells.Taken together,our study provided new insights into developing multi-stimuli responsive intracellular drug release systems for synergistic tumor cell therapy.
